摘 要:目的 探讨中药方剂抗敏合剂 (即过敏煎 )对致敏小鼠血清IgE的影响。 方法 用卵蛋白对小鼠进行致敏和气道激发 ,对照组给予等量PBS。 5周龄昆明小鼠分为对照组、致敏组、致敏 +药物干预组 ,在实验的第 1天 ,腹腔内注射卵蛋白 10 0mg/只 ,进行致敏 ,药物干预起始于致敏后第 7天 ,每天予药物胃管灌入 1次 ,分低、中、高 3个剂量组。于致敏后 7、10、14、17d即药物干预后 0、3、7、10d取材 ,观察各组血清IgE的动态变化 ;同样各组小鼠中的一部分 ,于致敏后第 14天进行卵蛋白气道激发 ,激发后 2 4~ 4 8h进行取材 ,观察各组小鼠血清IgE的改变。结果 腹腔注射大剂量卵蛋白可使小鼠血清IgE水平增高 ,对其药物干预可使IgE水平降低 ,中、高剂量组的作用尤其明显 ;气道激发后 ,血清IgE水平进一步增高 ,而这种增高在中、高剂量药物干预组均不明显。结论 抗敏合剂可降低致敏小鼠血清IgE水平 ,从而降低致敏小鼠变态反应敏感性。Objective To investigate the effectiveness of kangminheji in suppressing IgE production in a murine model of allergy developed by using ovalbumin (OVA) antigen as an allergen. Me- thods Kunming mices were purchased from Laboratory Animal Research Center (Madical School of Suzhou Univensity) and housed in a specific pathogen-free facility.All mices were 5 wk old at the time of the experiments.Mice received primary immunization with i.p. injection of 100mg soluble OVA on the first day.On the 14th day,a part of mices were placed in a Plexiglas chamber (10 cm×20 cm×20 cm) and exposed to aerosolized 1% ovalbumin for 30 min on two occasions 4 h apart.PBS (i.p. and aerosolized) was administered to mice on a similar scheduld as in the negative controls.Ag-sensitized (OVA-sensitized and OVA-challenged) mice were treated by oral Kangminheji from the 7th day after systemic sensitization following 3 regimens,designated high(0.8g),middle(0.4g),low(0.2g).The efficacy of this immunization/sensitization protocol is shown by serum IgE concentration.IgE levels in serum were measured at various time-points. Results Mice immunized intraperitoneally showed high titers of IgE 1 week after primary sensitization,there are higher titers of IgE after aerosol challenge of mice with ovalbumin.Mice that were treated with kangminheji in vivo after intraperitoneal immunization with OVA had significantly lower IgE levels than that in the NS-treated group. Conclusion These data suggest that kangminheji is useful in suppressing the production of IgE antibodies without toxic side effects.Thus kangminheji may be useful in suppressing the development of allergic sensitization in a murine model of allergy.
