摘 要:本文提出了一个借钛-4,5-二溴苯基荧光酮(DBPF)-Tween-80与蛋白质显色反应光度测定微量尿蛋白的高灵敏方法。在pH2.9,血清蛋白能与Ti-DBPF-Tween-80形成蓝色络合物,最大吸收波长为605nm。加入小量乙醇能提高方法的灵敏度和稳定性,牛血清蛋白含量在0~150μg/25ml范围内服从Beer定律。尿液中其它成分不干扰测定。用拟定的方法测定人尿蛋白,获得满意结果。A highly sensitive and convenient spectrophotometric method is described for the determination of urinary protein with 2, 6,7-trihydroxy-9-(4, 5-dibromophenyl)fluorone (DBPF)-titanium(Ⅳ) complex. At pH 2. 9 , the bovine serum albumin (BSA) forms a blue complex with Ti-DBPF-Tween-80 complex after heating at 40-45℃ for 15 min. The absorption maximum of the complex is at 605 nm. Small amounts of ethanol can give sensitive and stable result, other composition in urine does not interfere with determination, and Beer's law is obeyed in the range of 0 to 150 μg/25 ml for BSA. The human urine sample has been measured directly by the proposed method with satisfactory results.
