Experimental Study on Endotoxin—induced Damage to the Stria Vascularis of Inner Ear in Guinea Pigs

摘  要:将内毒素注入豚鼠内耳的外淋巴腔,并以注入生理盐水作对照,观察注射前后血管纹超微结构变化。结果对照组无显著改变。实验组血管纹边缘细胞的膜性细胞器(线粒体、内质网等)严重破坏,细胞间隙增宽,溶酶体增多,№^+-K^+-ATP 酶反应产物减少。作者认为内毒索引发血管纹严重损害,使耳蜗的水与离子代谢紊乱,破坏内耳生化环境并导致耳蜗能源衰竭,是中耳炎并发感音神经性聋的主要原因。The incidence of sensorneural hearing loss(SNHL)in association with otitis media is much higher than it was expected.This study presents animal experiments on toxic effect of endotoxin(LPS)in the inner ear with special reference to the stria vascularis.LPS was injected into the perilymphatic space of the inner ear for albino guinea pigs.Only normal saline was given for the control group.Electron microscopic evaluation on the stria vascularis was done before and 1,3,5 days after injection.There was little change observed in control ears.However,the stria vascularis manifested severe damages mainly in its organelles including the mitochondria and endoplasmic reticulum,especially in marginal strial cells of LPS ears.The mitochondria were swollen with their crests in disorder,broken or vanished.The endoplasmic reticulum was dilated and intercellular spaces widened.The lysosomes were increased and enzymal reactants of Na^+-K^+-ATPase diminished.It is concluded that LPS—induced ul- trastructural and enzymal damages to the stria vascularis provide presumptive ion inbalance in the inner ear.This results in changing endolymph composition and failure of energy supply to the Corti organ. That is responsible for the pathogenesis of SNHL in otitis media.
