摘  要:目的:观察皮下注射肾上腺髓质素(adrenomedullin,ADM)对大鼠高血压的影响。方法:一氧化氮合酶抑制剂左旋硝基精氨酸诱导大鼠高血压,大鼠皮下注射脂质体携带的ADM治疗后,测定血浆亚硝酸盐含量和离体胸主动脉组织一氧化氮合酶的活性。结果:大鼠皮下每天注射左旋硝基精氨酸连续4周,血压明显升高,并且显著抑制大鼠胸主动脉一氧化氮合酶的活性犤对照组为(1.70±0.13)nmol/(g·min),空白脂质体组为(1.10±0.34)nmol/(g·min),低剂量ADM组为(1.23±0.36)nmol/(g·min),高剂量ADM组为(1.50±0.32)nmol/(g·min),n=6,P<0.05犦和减少血浆中亚硝酸盐的含量犤对照组为(21.60±2.30)mmol/L,单纯脂质体组为(9.65±2.11)mmol/L,低剂量ADM组为(13.35±3.20)mmol/L,高剂量ADM组为(18.45±4.00)mmol/L,n=6,P<0.001犦。皮下注射ADM浓度依赖性地减轻了左旋硝基精氨酸导致的高血压的程度;提高大鼠胸主动脉一氧化氮合酶的活性和增加血浆中亚硝酸盐的含量(P<0.05)。结论:ADM通过一氧化氮合酶/一氧化氮途径降低血压。AIM:To investigate the effects of adrenomedullin(ADM) injected subcutaneously on rat hypertension. METHODS:Hypertension of rat was induced by Nw nitro L arginine(L NNA,nitric oxide synthase inhibitor) L NNA.After the rats were injected subcutaneously with ADM carried by liposomes,the content of plasma nitrite salt and the nitricoxide synthase(NOS)activity,that isolated from the rat aorta were measured. RESULTS:After the rats were injected subcutaneously each day with L NNA for 4 weeks,the blood pressure of rats were significantly elevated,the NOS activity of rat thoracic artery was obviously inhibited[(1.70±0.13) nmol/g per minute] in control group,(1.10±0.34) nmol/g per minute in blank liposomes group,(1.23±0.36) nmol/g per minute in low dosage ADM group,(1.50±0.32) nmol/g per minure in high dosage ADM group,respectively,n=6,P< 0.05],and the content of plasma nitrite was reduced,and the content of plasma nitrite was reduced[(21.60?à2.30£(c)mmol/L in control group,!?.65?à2.11£(c)mmol/L in liposomesa2only group,£¨13.35?à3.20£(c)mmol/L in low dosage ADM group,£¨18.45?à4.00£(c)mmol/L in high dosage ADM group,respectively,n=6, P< 0.001].The concentration of ADM injected subcutaneously reduced dependently the degree of hypertension induced by L NNA and increased the NOS activity of rat thoracic artery and the content of plasma nitrite salt(P< 0.05).CONCLUSION:ADM drops hypertension through a NOS/NOdependent way.
