护肾固精方对系膜细胞增殖及转化生长因子β_1的影响

摘　　要：目的 :观察护肾固精方 (HushenGujingFang ,HSGJF)对脂多糖 (LPS)诱导的大鼠肾小球系膜细胞 (mesangialcells ,MsC)增殖及其分泌转化生长因子 β1(TGFβ1)的影响。方法 :采用体外培养大鼠MsC和血清药理学方法 ,分为HSGJF血清组 (高、中、低 3个剂量亚组 )、LPS病理组及空白组 ,采用MTT法和酶联免疫吸附 (ELISA)法检测MsC增殖和TGFβ1含量。结果 :HSGJF血清对MsC无明显细胞毒性作用。 5 0 ,1 0 0 ,2 0 0ml/L浓度的血清在作用 2 4 ,4 8h后与正常大鼠血清比较 ,能明显抑制大鼠MsC增殖 (P <0 .0 5或P <0 .0 1 )。MsC经LPS刺激后 ,TGFβ1蛋白含量明显升高。而 3种不同浓度的含药血清均能在一定程度上对抗LPS诱导的MsC分泌TGFβ1的增加 ,这种抑制作用随用药时间及药物浓度的升高而增强。结论 :HSGJF对肾小球MsC增殖有明显抑制作用 ,并能显著干预活化的肾小球MsC分泌细胞因子 。Objective:To study the effects of the Hushen Gujing Fang (HSGJF),a traditional Chinese prescription, on the proliferation of the glomerular mesangial cells (MsC) and the secretion of transforming growth factor β 1 (TGFβ 1). Method: The MsC were cultured in vitro, and the seropharmacology method was employed. The cells were divided into serum groups (containing high, middle and low dosage of HSGJF, respectively), LPS group and control group. The proliferation of MsC and the secretion of TGFβ 1 were detected by MTT assay and ELISA respectively. Result: HSGJF had no significantly cytotoxic effects on the MsC.The density of HSGJF containing serum between 50 ml/L and 200 ml/L could obviously inhibit the proliferation of MsC after 24 or 48 hours compared to control group. After the MsC was stimulated by LPS, the level of TGFβ 1 protein increased significantly. The high, middle and low dosages of HSGJF containing serum all could antagonise the LPS induced increment of TGFβ 1. The inhibitory effects increased time and dose dependently. Conclusion: HSGJF has significantly inhibitory effects on the proliferation of MsC, and inhibits the cytokine secretion of activated glomerular MsC, thus it has the immunoregulation effects.
