双龙接骨丸对成骨细胞增殖和细胞周期的影响

摘　　要：目的探讨中药复方制剂双龙接骨丸对成骨细胞增殖和细胞周期的影响。方法双龙接骨丸生药给大鼠灌胃,1次/d,共7d。于最后1次灌胃后2h处死,制备含药血清。以原代培养的大鼠成骨细胞为实验对象,采用不同浓度的含药血清刺激成骨细胞,四氮唑蓝法(MTT)检测细胞的增殖情况,流式细胞仪分析细胞周期的构成。结果①双龙接骨丸30mL/L含药血清组细胞增殖率与空白对照组相比,差异无显著性意义(t=1.06,P>0.05);50,100,250,500,750,1000mL/L含药血清组均可明显促进细胞增殖,与对照组相比,差异有显著性意义(统计学数值分别为t=2.53,P<0.05;t=3.09,3.32,3.54,3.87,4.12,P<0.01)。②细胞周期构成分析发现,100mL/L双龙接骨丸含药血清增加S期细胞比率,促进细胞进入分裂增殖期。结论双龙接骨丸含药血清能有效促进成骨细胞的增殖。AIM:To explore the influence of traditional Chinese compound shuanglong jiegu wan on the proliferation and cell cycle of osteoblasts.METHODS:Rats were treated with shuanglong jiegu wan by gastric perfusion,once a day for 7 days,and then the rats were killed 2 hours after the last gastric perfusion for preparation of serum containing shuanglong jiegu wan.The primarily cultured osteoblasts of rats were stimulated with the serum containing shuanglong jiegu wan of different concentration.The cell proliferation rate was assayed by using the methyl thiazoly tetrazolium(MTT) method,and then the cell cycle was determined with the flow cytometry technique.RESULTS:①The cell proliferating rate in the serum containing shuanglong jiegu wan of 30 mL/L group was not significant different from that in the blank control group(t=1.06,P >0.05), while those in the serum containing shuanglong jiegu wan of 50,100,250,500,750 and 1 000 mL/L groups were obviously accelerated,and significantly different from that in the control group(t=2.53,P< 0.05;t=3.09,3.32,3.54,3.87 and 4.12,P< 0.01).②The analysis on the composing of cell cycle showed that the serum containing shuanglong jiegu wan of 10 mL/L could increase the cell ratio at S phase and promote the cells into the splitting proliferating phase.CONCLUSION:The serum containing shuanglong jiegu wan can effectively promote the proliferation of osteoblasts.
