摘 要:目的对碱性成纤维细胞生长因子(bFGF)与重组人骨形态发生蛋白-2(rhBMP-2)诱导成骨能力的量效关系进行研究。方法280只BALB/c小鼠采用掷硬币方法随机分为8组,每组35只,实验组rhBMP-2的剂量均为0.5mg,bFGF的剂量分别为0,2,10,40,80,300,500活性单位(IU);设立单纯牛松质骨载体为对照组。按实验设计,分别将rhBMP-2混悬液及bFGF-PVP溶液与牛松质骨载体充分混匀,负压下真空抽吸,冻干。将复合植骨材料植入小鼠右侧股部肌袋内,各组分别于术后3,5,7,9,14,21d共6个时间点取材,进行组织学、X线分析以及钙含量测定,观察各组的诱导成骨情况。结果①bFGF剂量为10~80IU时,间充质细胞增殖、分化,软骨细胞及新生骨形成均早于其他各组,成骨量多于其他各组,组织钙含量明显高于其他各组(t=4.44,P<0.05),提示此剂量bFGF使rhBMP-2的诱导成骨能力明显增强。②bFGF剂量为0,2,300IU时,各组在间充质细胞增殖、分化,软骨细胞及新生骨形成以及组织钙含量方面差异均无显著性意义(t=0.73,P<0.05),提示此剂量bFGF对rhBMP-2的诱导成骨能力无明显影响。③bFGF剂量为500IU时,术后21d,仅1例标本可见散在的极少量新骨形成,其组织钙含量明显低于其他各组(t=6.15,P<0.05),提示此剂量bFGF明显抑制了rhBMP-2的诱导成骨能力。④术后21d,单纯松质?AIM:To study the dose dependent stimulation of osteoinduction by recombinant human bone morphogenetic protein 2(rhBMP 2) enhanced by basic fibroblast growth factor(bFGF).METHODS:Totally 280 Balb/c mice were divided into 8 groups randomly by throwing coin,with 35 mice in each group.The dosage of rhBMP 2 was 0.5 mg and the dosage of bFGF was 0,2,10,40,80,300,500 active unit(IU) respectively;and the simple calf spongy bone carrier was treated as the control group.According to the experiment design,rhBMP 2 suspension,bFGF PVP solution and carriers were mixed completely before vacuum suction and freeze drying.Compound bone grafting materials were implanted into the right thigh pouches of the mice and samples were extracted respectively 3,5,7,9,14,21 days after the implantation.Then histological and X ray analysis were taken and calcium content was measured in order to observe the osteoinduction and osteogenesis of each group.RESULTS:In 10-80 IU bFGF groups,the proliferation and differentiation of mesenchymal cells,formation of chondrocytes and new bones were all earlier than those in other groups;The amount of osteogenesis was greater than that in other groups;and the calcium content was obviously higher than that in other groups(t=4.44,P< 0.05).These indicated that bFGF of these dosage could obviously enhance the osteoinduction and osteogenesis by rhBMP 2.In 0,2,300 IU bFGF groups,there was no significant difference about the proliferation and differentiation of mesenchymal cells,formation of chondrocytes and new bones in each group(t=0.73,P >0.05).These suggested that bFGF of these dosage had no obvious influence on the osteoinduction and osteogenesis by rhBMP 2.In 500 IU bFGF group,only few scattered new bones were seen in just one sample 12 days after the implantation and the calcium content was obviously lower than that in other groups(t=6.15,P< 0.05).These suggested that bFGF of this dosage could obviously inhibit the osteoinduction and osteogenesis by rhBMP 2.Twelve days after the implantation,no new bones were
