摘 要:背景磷酸三钙陶瓷人工骨的主要成分与骨基质中的无机成分相似,可作为填充材料为新骨形成提供支架,发挥骨传导作用。但单纯使用磷酸三钙不具有骨诱导能力,因此提高人工骨的骨诱导能力成为了人们研究的重点。目的观察鼠成骨细胞在体外与磷酸三钙陶瓷和钙离子混合培养时的生长情况,研究磷酸三钙和钙离子对成骨细胞的影响。设计完全随机对照的实验研究。地点和对象实验地点在华中科技大学同济医学院附属协和医院骨科实验室完成,研究对象为新生大鼠颅骨成骨细胞。干预用酶消化法分离培养新生大鼠颅骨成骨细胞,通过扫描电镜对生长于磷酸三钙陶瓷材料上的成骨细胞进行观察,利用[3H]-TdR掺入法检测磷酸三钙对成骨细胞的影响。用体外不同浓度的钙离子(2.5~10mmol/L)作用于成骨细胞,通过增殖细胞核抗原(proliferatingcellnuclearantigen,PCNA)免疫组化分析测定钙离子对成骨细胞增殖的影响。主要观察指标观察实验组和对照组中成骨细胞的生长情况。结果成骨细胞不仅很好地贴附于陶瓷表面,而且细胞的增殖活性未受到抑制。钙离子对体外培养的成骨细胞的增殖无负效影响。结论TCP陶瓷人工骨与成骨细胞有良好的生物相容性,对成骨细胞的生长有一定的促进作用。BACKGROUND:The main component of the artificial bone of tricalcium phosphate(TCP) porcelain is similar to the inorganic component in bone matrix and can be used as support frame for bone formation.But TCP only can not induce bone formation.So it has become the focus of study to improve the bone inducing ability of artificial bone.OBJECTIVE:To observe the growth of rat osteoblast in vitro when cultured with TCP porcelain and Ca2+and to study the effect of TCP porcelain and calcium cation on osteoblast.DESIGN:A completely randomized controlled experiment was conducted.SETTINGS and PARTICIPANTS:The experiment was conducted in Osteoplastic Laboratory of affiliated Union Hospital of Tongji Medical College,Huazhong University of Science and Technology with the subject of cranial osteoblasts of newborn rat. INTERVENTION:The cranial osteoblasts of newborn rat were separated and cultured with enzyme digestion method.The osteoblasts on TCP porcelain were observed through scanning electron microscope and the effect of TCP porcelain on osteoblasts was examined with TdR corporation method.The osteoblasts were cultured in different concentrations of Ca2+(2.5-10 mmol/L) whose effect on the proliferation of osteoblasts was measured with PCNA(proliferating cell nuclear antigen)immunohistochemical assay.MAIN OUTCOME MEASURES:The growth of osteoblasts in both experiment groups and control group.RESULTS:Not only the osteoblasts adhere well to the surface of porcelain but their proliferation activity is also not inhibited.No negative effect of Ca2+is observed on the proliferation of osteoblasts cultured in vitro.CONCLUSION:The artificial bone of TCP porcelain is biocompatible with osteoblast and can promote the growth of osteoblast to a certain degree.
