当归补血汤含药血清抗动脉粥样硬化的机理研究——一对氧化修饰低密度脂蛋白趋化单核细胞作用的影响

摘　　要：[目的]观察当归补血汤含药血清对氧化修饰低密度脂蛋白(oxidized low-density lipoproteins,OxLDLs)趋化单核细胞(monocytes,MC)作用的影响。[方法]新西兰兔6只,随机分为当归补血汤组、单味当归组和单味黄芪组,以相应药物灌胃3d,2次/d,第3天给药后1h心脏取血分离含药血清,并设自由趋化组(正常组)和OxLDLs组(模型组)作对照,用趋化小室微孔滤膜法检测各组OxLDLs趋化MC的数目及成像。[结果]当归补血汤组中OxLDLs趋化MC数目比模型组减少(P<0.01),而黄芪含药血清和当归含药血清组MC数目与模型组无显著性差异。[结论]当归补血汤全方可抑制OxLDLs对MC的趋化性,从而对MC向血管内皮下聚集而导致泡沫细胞的形成并进而引起的动脉粥样硬化起一定的抑制作用;而当归和黄芪单独使用未见此效应。[Objective] To investigate the influence of serum containing Danggui Buxue Decoction (DBD) on chemotaxis of oxidized low-density lipoproteins ( OxLDLs) for monocytes ( MC). [ Methods ] Six New Zealand rabbits were randomized to DBD group, Radix Angelicae Sinensis (RAS) group and Radix Astragali seu Hedysari (RASH) group. The rabbits were treated by gastric infusion of the above drugs respectively, bid for three days. On the third day, the serum containing drugs was obtained by heart blood sampling one hour after administration. Free chemotaxis group (normal control group) and OxLDLs group ( model group) were established for control. The number and figure of OxLDLs - chemotactic MC were observed by micropore filter method with chemotactic chamber. [Results] OxLDLs -chemotactic MC number in DBD group were lower than those in model group (P < 0.01). The difference of MC number was insignificant between RAS group and RASH group. [Conclusion] DBD has an inhibitory effect on chemotaxis of OxLDLs for MC, thus counteracts the accumulation of MC in vascular endodermis, inhibits the formation of foam cells and blocks the pathological progress of atherosclerosis. However, RAS or RASH alone has no such effect.
