类M蛋白对HEp-2细胞的黏附作用

摘　　要：采用通用的模式细胞HEp 2作黏附及黏附抑制试验 ,研究马链球菌兽疫亚种ATCC35 2 4 6株类M蛋白的黏附作用。结果表明 ,ATCC35 2 4 6株可以黏附HEp 2细胞。用重组表达的类M蛋白鼠抗血清处理ATCC35 2 4 6 ,可抑制其对HEp 2细胞的黏附 ,最大抑制率达 81 4 % ;而ATCC35 2 4 6的全菌鼠抗血清在 1∶80 0时能完全抑制它对HEp 2细胞的黏附。此外 ,热酸提取的ATCC35 2 4 6株的类M蛋白和重组表达的类M蛋白可不同程度地抑制该菌株的黏附 ,前者在 16 0μg·mL-1有最大的黏附抑制率 ( 4 0 % ) ,而后者在 6 0 μg·mL-1的黏附抑制率可达最高值 ( 2 3% )。结果显示 ,类M蛋白是ATCC35 2 4 6株的黏附素之一 。HEp 2 cells,a type of common cell model,was used in the adhesion and adhesive inhibition experiments to evaluate the adhesion role of M like protein from Streptococcus equi subsp. zooepidemicus strain ATCC35246.The adhesion results showed that ATCC35246 could adhere HEp 2 cells and the number of adhesive bacteria were 72 per cell.Pretreatment of ATCC35246 with anti mouse serum of purified recombinant M like protein could reduce its adhesion activity by 81 4%,while anti mouse serum of ATCC35246 at titer of 1∶800 could completely block its adhesion.In addition,hot acid extracted and recombinant M like protein reduced the adhesion activity of ATCC35246 at different degree.The former had its maximum inhibition capability at concentration of 160 μg·mL -1 ,and the percentage of adhesive inhibition approximated to 40%.While the highest percentage of adhesive inhibition of the latter was 23% at its concentration of 60 μg·mL -1 .It demonstrated that M like protein might be an adhesin of ATCC35246 and play important role in the adhesion of Streptococcus to host cells.
