摘  要:白细胞介素 1β(IL 1β)及受体表达增加可见于脑缺血、外伤和退行性病变等多种病理情况,而对IL 1β作为中枢神经系统信号物质的作用及其机制知之甚少.本实验采用全细胞电压钳记录,研究了不同浓度的IL 1β对急性分离大鼠海马锥体细胞Na+电流的调制作用.实验结果如下:1)IL 1β(0.1ng/mL)使Na+电流峰值增加,但对离子通道动力学无显著影响;2)IL 1β(10ng/mL)对海马神经元电压门控钠通道具有抑制作用,并使Na+通道开放阈值提高.实验结果提示:1)中枢神经系统中存在的IL 1β基础分泌可以通过电压门控Na+离子通道参于神经元兴奋性调节;2)IL 1β水平的异常增加对Na+通道具有抑制作用,对防止损伤导致的神经元去极化具有积极意义.Elevated levels of Interleukin 1β(IL-1β)are present in the brain tissue during pathological processes. In the present study we examined the effects of recombinant murine IL-1βon voltage gated sodium channel of acutely isolated rat hippocampal neurons using the whole-cell patch-clamp technique. The results indicated that IL-1β voltage-dependently increased sodium currents at physiological concentration (0.1 ng/mL) without alteration of the parameters of channel kinetics.While IL-1βat pathophysiological concentration (10 ng/mL) depressed the sodium currents in a voltage-dependent manner and shifted the I-V curve of sodium currents towards positive potential without changing the slope. The steady state inactivation curve was not affected by IL-1βat both concentrations. The results suggest that 1)the basal level of IL-1βin central nervous system is involved in the modulation of neuronal excitability through voltage gated sodium channel ;2) elevated level of IL-1β may play a protective role in pathological processes by inhibiting neuron depolarization caused by damage.
