摘 要:目的:探讨不同浓度的米非司酮和利洛司酮对异位子宫内膜间质细胞血小板衍生生长因子(PDGF)和血管内皮生长因子(VEGF)表达和体外增殖的影响?方法:应用免疫组织化学法检测异位间质细胞PDGF表达?应用ELISA检测间质细胞培养基上清液中VEGF浓度?应用RT-PCR方法检测异位间质细胞PDGF?VEGF的表达?结果:米非司酮组0?1×10-6?1×10-5mol/L 浓度段,PDGF阳性率分别为65%?48%?34%(P<0.05);VEGF量分别为(561.8±22.4)?(417.4±24.8)?(319.4±24.9) ng/L (P<0.01)?利洛司酮组0?1×10-6?1×10-5mol/L 浓度段,PDGF阳性率分别为65%?45%?31%(P<0.05);VEGF量分别为(561.8±22.4)?(327.8±24.7)?(251.0±18.8) ng/L (P<0.01)?PDGF和VEGF蛋白和mRNA表达,米非司酮组高于利洛司酮组,对照组依次高于抗孕激素1×10-6和1×10-5 mol/L组?结论:米非司酮及利洛司酮以剂量依赖性方式抑制异位子宫内膜间质细胞的体外增殖, 其作用与降调细胞生长因子PDGF和VEGF的表达有关?Objective: To investigate the effects of mifepristone and lilopristone on the expression of PDGF, VEGF and proliferation of the ectopic stromal cells in vitro. Methods: The expression of PDGF of ectopic stromal cells were determined by immunohistochemistry. The concentration of VEGF in the supernatant of the media was assayed by ELISA. The expression of PDGF and VEGF mRNA of ectopic stromal cells was detected by RT-PCR. Results: The expression of PDGF was 65%,48% and 34%(P<0.05) respectively in the group of 0, 1×10-6 and 1×10-5 mol/L of mifepristone, and 65%,45% and 31%(P<0.05) respectively in the group of 0, 1×10-6 and 1×10-5 mol/L of lilopristone. The quantity of VEGF in the supernatant was 561.8±22.4, 417.4±24.8 and 319.4±24.9 ng/L (P<0.01) respectively in the group of 0, 1×10-6 and 1×10-5 mol/L of mifepristone, and 561.8±22.4, 327.8±24.7 and 251.0±18.8 ng/L (P<0.01) respectively in the group of 0, 1×10-6 and 1×10-5 mol/L of lilopristone. The expression levels of PDGF mRNA and VEGF mRNA of ectopic stromal cells in both antiprogestin cultutred groups were lower than that of control group; the expression levels of antiprogestin 1×10-6 mol/L group was higher than that of 1×10-5 mol/L group. The expression level in mifepristone group was higher than that of lilopristone group. Conclusion: Antiprogestin mifepristone and lilopristone could significantly suppress the proliferation of ectopic stromal cells in the dose-dependent fashion in vitro. The mechanisms were associated with down-regulation of the expression of PDGF and VEGF. [
