摘 要:目的:观察阴虚阳亢模型小鼠Th1/Th2两类细胞因子的漂移现象及中药左归丸的逆转作用?方法:采用甲状腺素合并番泻叶建立阴虚阳亢小鼠模型,并用中药左归丸对模型的建立进行干预(生理盐水做对照),RT-PCR法分别检测小鼠脾脏单个核细胞Th1类细胞因子(IFN-γ?IL-2)?Th2类细胞因子(IL-4?IL-10)的表达及中药左归丸的逆转作用?结果:正常小鼠可以同时表达IFN-γ?IL-2?IL-4?IL-10,处于Th1/Th2平衡状态;模型小鼠在表现明显的阴虚阳亢体征的同时,其Th1/Th2两类细胞因子mRNA的表达显著降低(P<0.01),但IFNγ/IL-10的比值明显升高(P<0.01),Th1类细胞因子处于相对优势状态,中药左归丸可显著提高IL-10的转录水平,使IFNγ/IL-10比值回落(P<0.05)?结论:中医阴虚阳亢小鼠处于Th1相对优势状态,中药左归丸可重建Th1/Th2的平衡状态?Objective: To observe the profile of Th1/Th2 type cytokine in yin-deficient model mice and the effects of the decoction of ZUOGUIWAN, which is the traditional prescription of yin-enrichment Chinese herbs. Methods: The yin-deficient model was established by treatment of BALB/c mice with Thyroxin combined with Senna leaf (ig), meanwhile, a part of the model mice were treated with ZUOGUIWAN (compared with normal saline). When the model was finished, or after it was treated with ZUOGUIWAN, the transcripts of IFN-γ, IL-2 (Th1) and IL-4, IL-10 (Th2) of splenocyte were tested by RT-PCR. Results: Normal mice expressed IFN-γ, IL-2 and IL-4, IL-10 simultaneously, with a Th1/Th2 balance status (Th0). The mRNA production of IFN-γ, IL-2, IL-4, IL-10 was decreased in yin-deficient mice (P<0.01), but the ratio of IFN-γ/IL10 was significantly increased (P<0.01). Th1 type cytokines were relatively in dominant status and the treated model mice with ZUOGUIWAN were characterized with descent tendency of IFN-γ/IL-10 through the up-regulation of IL-10(P<0.05). Conclusion: The results suggested that yin-deficient status may be associated with the relative Th1 predominant response and the ZUOGUIWAN can reestablish the new balance of Th1/Th2. [
