摘  要:目的:分析雌激素对大鼠血管平滑肌细胞(VSMCs)增殖及Id3α基因表达的影响,探讨雌激素抑制VSMCs增殖的分子学机制?方法:培养Wistar大鼠血管平滑肌细胞,经17β-雌二醇 (17β-E2)处理24 h后,采用MTT比色法测定细胞增殖水平,并提取细胞总RNA,经甲醛变性凝胶电泳后行Northern blotting分析,观察雌激素对血管平滑肌细胞Id3α基因mRNA表达的影响?结果:经17β-E2处理的VSMCs增殖受到抑制,并呈现浓度依赖趋势, Id3α基因mRNA表达明显增强?结论:雌激素抑制VSMCs增殖,对Id3α基因表达的上调作用可能是其发挥抑制作用的分子机制之一?Objective: To elucidate the effect of estrogen on the proliferation of vascular smooth muscle cells (VSMCs) and the involved possible molecular mechanism by analyzing the influence of estrogen on mRNA expression of Id3α in rat VSMCs. Methods: After pretreated with 17β-E2 (17β-Estradiol) for 24 hours, the number of cultured rat VSMCs was determined by MTT colorimetric method , and the expression of Id3α mRNA was analyzed by Northern blotting analysis. Results: Compared with the cells unpretreated with 17β-E2, the proliferation of VSMCs treated with 17β-E2 (1~100nmol/L) was inhibited in a concentration-dependent manner, and the mRNA expression of Id3α was significantly higher. Conclusions: Estrogen inhibits the proliferation of VSMCs and the up-regulation of Id3α gene expression may represent part of the involved molecular mechanisms. [
