摘 要:目的 :观察辛伐他汀 (Sim)抑制大鼠右心室肥大的作用。方法 :一次性皮下注射野百合碱 (MCT) 5 0mg·kg-1 ,引起大鼠肺动脉高压和右心室肥大。 2周后 ,分组灌胃给予Sim 1 .5 ,3.0mg·kg-1 ·d-1 或纯化水 (对照组 ) ,连续治疗 1 4d。测定各组大鼠的平均肺动脉压、右心室和左心室 +室间隔的重量比 [R/(L +S) ]、右心室组织的蛋白和羟脯氨酸含量 ,并观察右心室和肺的病理形态。结果 :Sim 3.0mg·kg-1 ·d-1 给药 1 4d ,使R/(L +S)、蛋白质和羟脯氨酸含量分别比对照组大鼠减少 35 %,6 2 %,38%(P <0 .0 1 )。但对MCT引起的大鼠肺动脉压升高和肺组织形态改变无影响。结论 :Sim对MCT引起的大鼠右心室肥大的进程具有抑制作用。OBJECTIVE To observe the effect of simvastatin (Sim) on right ventricular hypertrophy induced by monocrotaline (MCT) in rats. METHODS Pulmonary hypertension and right ventricular hypertrophy in rats were induced by a single subcutaneous injection of MCT(50 mg·kg -1). Two weeks after injection MCT, the rats were treated with Sim ( 1.5 and 3.0 mg·kg -1·d -1) or distilled water (control) for fourteen days. Then the mean pulmonary arterial pressure, the ratio of right ventricle weight to left ventricular-plus-septum weight [R/(L+S)], lung and hydroxyproline content of right ventricle were measured. Morphology of lung and right ventricle was observed. RESULTS After the rats were treated with Sim 3.0 mg·kg -1·d -1, R/(L+S), protein and hydroxyproline content of right ventricle decreased by 35%, 62%, and 38%, respectively compared with control (all P< 0.01). Sim had no effect on pulmonary hypertension and morphologic changes of pulmonary in MCT rats. CONCLUSION Sim inhibited the development of right ventricular hypertrophy in rats of pulmonary hypertension induced by MCT.
