乙型肝炎慢性化与CⅡTA基因启动子区多态性的相关性

摘　　要：目的 :探讨CⅡTA基因启动子区的多态性与乙型肝炎慢性化的相关性 .方法 :用聚合酶链反应 单链构象多态性 (PCR SSCP)及DNA测序方法对 1 0 2例慢性乙型肝炎患者和80例正常对照者外周血白细胞基因组DNA的CⅡTA基因Ⅰ ,Ⅲ及Ⅳ 3个启动子的基因多态性进行分析 .结果 :1 0 2例患者和 80例正常人CⅡTA基因的第Ⅰ ,Ⅲ及Ⅳ 3个启动子的PCR产物均呈一带型 ,未发现异常泳动 .结论 :PCR SSCP分析显示CⅡTA基因的第Ⅰ ,Ⅲ及Ⅳ 3个启动子在所研究的人群中不存在多态性 。AIM: To explore the relationship between polymorphism of CⅡTA gene promoters and the chronicity of hepatitis B. METHODS: The promoterⅠ,Ⅲ and Ⅳ of CⅡTA gene was analyzed, respectively with polymerase chain reaction single strand conformation polymorphism (PCR SSCP) in 102 patients with chronic hepatitis B and 80 normal controls. RESULTS: No abnormal migration was found in the PCR SSCP analysis of the two groups. CONCLUSION: No polymorphism in promoterⅠ,Ⅲ and Ⅳ of CⅡTA gene is found in the analyzed subjects. This results suggest that the promoter of CⅡTA gene may be a conserved domain.
