摘　　要：目的 :探讨去甲斑蝥素 (NCTD)对人胆囊癌细胞侵袭转移及其相关基因的抑制效应。方法 :培养人胆囊癌GBC SD细胞 ,应用Matrigel实验、过河实验、SABC方法检测NCTD对GBC SD细胞侵袭、运动和基质溶解相关基因蛋白明胶酶A即金属蛋白酶 2 (MMP2 ) ,金属蛋白酶 2组织抑制剂 (TIM P2 )表达的影响。结果 :NCTD在 5mg·L- 1时即可抑制GBC SD细胞的体外侵袭、运动能力 ,随药物浓度提高过膜细胞减少 ,过膜死亡细胞增多 ,过河时间延长(P <0 .0 1) ;GBC SD细胞MMP2 表达下降 ,TIMP2表达上升 ,MMP2 /TIMP2 比值下降 (P <0 .0 1)。结论 :NCTD在低浓度下即可抑制人胆囊癌细胞的体外侵袭、运动能力 ,其机制可能与抑制GBC SD细胞迁移运动和干扰GBC SD细胞MMP2 ,TIMP2 表达有关。AIM: To explore the influence of norcantharidin on invasion of GBC-SD cells in human gallbladder carcinomas and inhibitory effect of the related genes. METHODS: GBC-SD cells of human gallbladder carcinoma were cultured by cell culture technique. The Matrigel experiment and the crossing-river test were used to examine the inhibitory effect of norcantharidin on invasion of GBC-SD cells. The influence on expression of MMP_2 and TIMP_2 proteins of GBC-SD cells was determined by streptavidin-biotin complex method before and after the treatment with norcantharidin. RESULTS: The Matrigel experiment showed that norcantharidin began to inhibit the invasion of GBC-SD cells at the concentration of 5 mg·L -1 ,and it could completely inhibit the invasive action of GBC-SD cells at 40 mg·L -1 .The crossing-river test indicated the crossing-river time of GBC-SD cells treated with norcantharidin (40 mg·L -1 ) for 72 h became long significantly. The expression of MMP_2 gene proteins and the ratio of MMP_2/TIMP_2 of GBC-SD cells decreased significantly,and expression of TIMP_2 increased( P <0.01 ). CONCLUSION: Norcantharidin can inhibit the invasive capability of GBC-SD cells in human gallbladder carcinomas. The mechanism may correlate with inhibition of GBC-SD cell movement and influence of the expression of MMP_2 and TIMP_2 of GBC-SD cells.
