摘 要:本研究测定了石英粉尘对豚鼠肺泡巨噬细胞化学发光的影响,以二氧化钛粉尘为阴性对照粉尘,所采用的粉尘剂量分别为50、100和200μg/10~6细胞。结果表明,石英粉尘引起肺泡巨噬细胞化学发光强度随粉尘剂量增加而增强,有明显的剂量反应关系。石英组由低剂量到高剂量化学发光强度峰值分别为15.70、23.08及46.46(mV),对照组为10.25(mV)。而二氧化钛粉尘仅在200μg剂量组化学发光反应增强,峰值为18.83(mV)。值得注意的是,石英粉尘200μg剂量组在加尘后3分钟反应迅速达到高峰,而后随粉尘作用时间延长,发光强度明显减弱,至测定的60分钟,接近低剂量组水平。巨噬细胞化学发光的测定可以反映粉尘毒性大小和细胞的早期损伤,这对探讨尘肺的发病机理有重要意义。The influence of quartz on guinea pig alveolar macrophage oxidative metabolism was studied by a chemiluminescence (CD assay.Quartz caused a concentration-dependent enhancement of CL response.Thus, as the quartz at 50, 100 and 200μg/106cells concentration the CL response was augmented to 15.70, 23.08 and 46.460(mV) respectively (control: 10.25 mV). Titanium dioxide (TiO2) dust used as an inert control did not cause a significant CL response at low concentration.However, at higher concentration of 200μg/106 cells, CL response was 18.83(mV).It was very interesting that the CL response reached to the maximum value induced by quartz at a concentration of 200μg/106 cell in 3 minutes. Then CL intensity was decreased significantly with the prolongation of the exposure to quartz. And CL intensity reached to that of low concentration level in 60 minutes. The CL assay of macrophage can be used in the study on the toxicity of dusts and early damage of cell by silica.
