摘 要:了解邻苯二甲酸丁基苄酯 ( BBP)在肺匀浆中是否被代谢以及被代谢的情况 ,为更深入的肺脏毒性研究提供参考。于肺匀浆液中加入一定量 BBP后 ,在 0 min、10 min、2 0 min、40 min、60 min、80 min不同的温孵时间用乙酸乙酯灭活萃取剩余 BBP,处理后用高效液相色谱测定 BBP含量 ,再通过标准曲线计算出测定的 BBP含量 ,得出BBP在肺中被降解的量 ,计算得到不同时间 BBP在肺匀浆液中的降解百分率。研究结果表明 ,BBP在肺匀浆液中的线性范围为 7.2~ 179μmol/ m L,平均萃取回收率为 ( 97.11± 0 .98) %。 BBP在肺匀浆液中被降解 ,与时间呈一定线性关系 ;代谢上有雌雄差异 ,雌性在 40 min以前降解比雄性快 ,在雌性肺匀浆中的降解半衰期为 93 .65 min,在雄性肺匀浆中的降解半衰期为 70 min,但是雄性在整个过程中降解较快 ;采用乙酸乙酯进行灭活萃取 ,在本实验条件下没有检测到代谢产物 ,说明代谢产物极性较大 ,没有被乙酸乙酯萃取出来。The present paper aims to disclose the metabolic mechanism of BBP in lung homogenate and metabolic course in hope to prove the reliance of in-depth study on lung toxicity. To achieve the goal, the authors first of all added BBP in lung homogenate, and then, extracted BBP by ethyl acetate in 0, 10, 20, 40, 60 and 80 minutes, respectively. Since the High Performance Liquid Chromatography can help to detect the apex intense, it is possible to calculate the quantity of BBP in a standard curve and deduce the degradation quantity and ratio. Next, an ultraviolet detector can be adopted with OSD column with a mobile phase consisting of methanol-water (80∶20, V∶V), a 0.8 mL/min velocity of mobile phase and a 254 nm detective wavelength. The above method helps to produce a very nice linearity for BBP in the range 7.2-179 μmol/mL in the lung homogenate, with the mean recovery rate at about (97.11±0.98)%. The degradation of BBP will occur in the lung homogenate in a linear relation with the time. However, sex difference can also be found in this experiment with which the female's lung homogenate half life degradation being 93.65 minutes in contrast with male being 70 minutes. This indicates a faster degradation in female lung homogenate before 40 minutes during the whole degradation period. However, there were no chromatographic metabolites seen in this experiment to verify the metabolites are polar compounds with ethyl acetate applied to the extract of BBP from the lung homogenate.
