摘 要:目的 研究神经细胞突起生长诱向因子 (β Netrin)在大鼠脑内的基因表达。 方法 以地高辛标记的cR NA探针利用组织原位杂交技术检测β NetrinmRNA在出生后 30d大鼠脑内的表达及其分布。结果 (1)脑内β NetrinmRNA主要定位于神经元 ;(2 )脑内β NetrinmRNA阳性细胞广泛分布于大脑皮层、边缘系统、小脑、脑干等处的神经核团及嗅球中。其中 ,梨状前皮质、杏仁核群、海马各区锥体层、小脑浦肯野细胞、小脑内侧顶核及间置核、斜方体内侧核和嗅球僧帽细胞等部位阳性信号较强 ;中等强度的阳性信号见于额叶皮质、丘脑外侧背核等处。结论 原位杂交结果显示β NetrinmRNA在成年大鼠脑内神经元中有表达 ,进一步验证了β Netrin对脑内神经细胞突起的生长具有诱向作用。同时 ,由于β Netrin在大鼠嗅觉传导相关通路和海马、大脑皮质内表达量较高 ,提示βObjective To investigate the expression and distribution of the β-Netrin mRNA in the brain of adult rat.Methods In situ hybridization with the Digoxigenin-labeled cRNA probe. Results (1)The positive signal of β-Netrin mRNA was localized in the cytoplasm and processes of neuron; (2)The distribution of β-Netrin mRNA was mainly detected in the regions of cerebral cortex, limbic system, thalamus, cerebellum, olfactory bulb and some neural nucleus mass. The strong dense signal of β-Netrin mRNA was found in the layer of pyramidal cells of cerebral cortex, prepiriform cortex, amygdaloid nuclei, pyramidal layer of hippocampus, Purkinje cells, medial cerebellar nucleus and interposed cerebellar nucleus, medial mucleus of the trapezoid body and mitral cell layer of the olfactory bulb. The moderate dense signal was present in frontal cortex, several thalamic and hypothalamic nuclei, ventrolateral nucleus, laterodorsal thalamic nucleus such as laterodorsal thalamic nucleus, etc. Conclusion Results showed that β-Netrin is a very important gene in the brain of adult rat. Considering the function of β-Netrin in axon guidance, it could be inferred that there are still some active axon growth and guidance in the adult stage. Moreover, the high level expression of β-Netrin mRNA in the region of hippocampus and cerebral cortex suggested that β-Netrin may also take part in the process of learning and memory.
