摘  要:目的探讨实验性哮喘时神经生长因子(nervegrowthfactor,NGF)对降钙素基因相关肽(calcitoningenerelatedpeptide,CGRP)的调节机制。方法应用放射免疫分析方法检测卵蛋白致敏致喘豚鼠下呼吸道和内脏感觉传入部位CGRP含量的变化。结果哮喘组豚鼠气管、支气管、肺、C7~T5段脊神经节和相应节段脊髓后角、结状神经节及孤束核区CGRP含量(pg/g)分别为71.72±17.82,75.56±19.23,82.85±23.33,83.83±21.29,74.47±22.25,77.65±20.21和69.95±12.27,显著高于生理盐水对照组(48.06±12.32,53.36±15.22,62.39±20.25,67.76±20.29,60.63±14.45,65.54±17.27,59.33±21.22),差异有显著性意义(P<0.01)。在哮喘+NGF抗体组,上述部位内CGRP含量(pg/g)分别为35.25±8.76,42.26±10.96,58.89±18.83,62.67±22.24,56.55±16.69,60.34±17.97和54.46±16.62,明显低于哮喘组,差异有显著性意义(P<0.01)。结论实验性哮喘时NGF可上调下呼吸道和感觉神经节等内脏感觉传入部位的CGRP水平,两者可能参与支气管哮喘的发病过程。AIM:To explore the regulatory mechanism of nerve growth factor (NGF) on calcitonin gene related peptide(CGRP) in experimental asthma.METHODS:Radioimmunoassay was used to determine the alteration of CGRP levels in the lower respiratory tract and visceral afferent sites while NGF was absent(inhalation of NGF antibody through nasal cavity) in the asthmatic guinea pig.RESULTS:The contents(pg/g) of CGRP in the trachea,bronchus,lung,C7-T5 spinal ganglia and correspondent spinal dorsal horn,nodose ganglia and solitary nucleus area in the asthmatic group were 71.72±17.82,75.56±19.23,82.85±23.33,83.83±21.29,74.47±22.25,77.65±20.21 and 69.95±12.27,respectively,significantly higher than those in the normal saline control group[(48.06±12.32),(53.36±15.22),(62.39±20.25),(67.76±20.29),(60.63±14.45),(65.54±17.27) and(59.33±21.22)pg/g](P< 0.01);while those in the asthmatic+NGF antibody group[(35.25±8.76),(42.26±10.96),(58.89±18.83),(62.67±22.24),(56.55±16.69),(60.34±17.97) and (54.46±16.62)pg/g] were obviously lower than those in the asthmatic group(P< 0.01).CONCLUSION:NGF can upregulate the contents of CGRP in the lower respiratory tract and visceral sensory sites of the experimental asthmatic guinea pig,both of which may be involved in the pathogenesis of asthma.
