补肾活血方药对老年小鼠睾丸的影响

摘　　要：目的补肾活血中药左归饮加减其作用针对肾虚这一衰老的基本病机,研究其对老年小鼠睾丸的影响,有助于揭示其抗衰老机制。方法昆明种雄性小鼠36只,其中老年小鼠(18～22个月)24只,随机分为老年实验组和老年对照组,12只4～5个月小鼠为青年对照组。以中药方剂左归饮加减的水煎液供实验组小鼠自由饮用。75d后,取各组小鼠睾丸进行组织学和细胞化学研究。结果老年实验组小鼠曲精小管增粗(152.08±9.25)μm与老年对照组(139.4±14.42)μm比较,差异有非常显著性意义(t=3.102,P<0.01);平均支持细胞数(12.03±3.72)与老年对照组(9.11±2.18)比较,差异有显著性意义(t=1.809,P<0.05);核仁增大;间质细胞核质比例(0.44±0.40)与老年对照组(9.11±2.18)比较差异有显著性意义(t=1.801,P<0.05);3β-羟基甾脱氧酶反应增强;增殖型精原细胞平均数(40.08±15.63)与老年对照组的(29.25±11.39)比较,差异有显著性意义(t=1.804,P<0.05)。结论以左归饮为主的补肾活血方药有调整下丘脑-垂体-性腺轴的分泌机能,促进睾丸的生精活动从而起到抗衰老的作用。AIM:The effect of the traditional Chinese medicine for tonifying idney and invigorating blood circulation named zuogui yin jiajian is aiming directly at the deficiency of kidney which is the basic mechanism of antiaging, the aim of this study was to investigate its influence on the testes of the aging mice and to explore its antiaging mechanism.METHODS:Totally 36 male mice of Kunming species were used in the study,and 24 aging mice(18-22 months old) of them were divided into old experimental group and old control group,the other 12 mice(4-5 months old) were selected as the young control group.The mice in the old experimental group received the treatment of free drinking of water decoction of zuogui yin,and after 75 days,the testes of mice in all groups were taken for histological and cytochemical studies.RESULTS:The seminiferous tubules became thicker in the old experimental group[(152.08±9.25) μm],very significantly different from that in the old control group[(139.4±14.42) μm](t=3.102,P< 0.01).The mean number of Sertoli cells in the old experimental group(12.03±3.72) was significantly different from that in the old control group(9.11 ±2.18)(t=1.809,P< 0.05).The nucleolues became larger.The nucleo cytoplasmic ratio of interstitial cells in the old experimental group(0.44±0.40) was significantly different from that in the old control group(9.11 ±2.18)(t=1.801, P< 0.05).The reaction of 3 beta hydroxy steroid dehydrogenase was strengthened and the mean number of proliferating spermatogonium(40.08±15.63) also increased significantly as compared with that in the old control group(29.25±11.39)(t=1.804,P< 0.05).CONCLUSION:The traditional Chinese herbal medicine mainly made of zuogui yin for kidney tonic and circulation of blood invigoration can regulate the secreting function of hypothalamic hypophyseal gonadal axis,and promote the spermatogenesis process on testes of aging mice so as to play its role against aging.
