摘  要:目的 探讨肌动蛋白细胞骨架稳定剂鬼笔槐酞 (Phalloidin)对机械牵张复合模拟缺血缺氧损伤心肌细胞肌球蛋白重链 (myosinheavychain ,MHC)mRNA表达的影响。方法 建立离体培养的心肌细胞机械牵张模型 ,并施加无糖缺氧刺激因素 ,模拟烧伤后缺血缺氧损害。采用RT PCR方法检测phalloidin对心肌细胞MHCmRNA表达的影响 ,并利用凝胶分析软件对PCR结果进行分析。结果  10 %牵张导致MHCmRNA由α型、β型表达均增加 ,且以 β型增加为主。模拟缺血缺氧时MHCmRNAα型减少 ,β型增多后下降。 10 %牵张 +模拟缺血缺氧时 ,α型、β型变化加剧 ,随刺激时间的延长转化加剧 ,12h后 ,α型、β型MHCmRNA表达均下调 (P <0 .0 1、P <0 .0 5 )。应用Phalloidin后 ,显著改善了 10 %牵张 +模拟缺血缺氧时α型向 β型的异常转化。 结论 机械牵张进一步加剧了模拟缺血缺氧对MHC基因表达由α型向 β型的异常转化及下调 。Objective To investigate the effects of phalloidin on MHC gene expression of cardiomyocyte treated by static stretch in combination with ischemia and hypoxia in vitro . Methods Cardiomyocytes were cultured in vitro and stimulated by mechanical stretch in combination with ischemia and hypoxia. Before and after addition of phalloidin into culture media, the changes of MHC gene expression was detected by RT PCR and analysis were carried out using gel image analysis software. Results Static mechanical stretch on cells resulted in an increase of both α and β MHC, but β MHC increased more obviously. In cells treated with ischemia and hypoxia, changes of MHC gene changing from α to β accelerated. At 12 h after treatment, the sum of α and β mRNA expressions were down regulated. When static mechanical stretch was added in cells treated with ischemia and hypoxia, the change from α to β exaggerated. The expression of the two genes decreased at 12 h after treatment. In phalloidin treatment group, MHC genes maintained normal expression to some extent in contrast with the former group. Conclusion Mechanical stretch combined with ischemia and hypoxia down regulates MHC gene expression in cardiomyocytes, which might be one of the factors leading to cardiac dysfunction during the early stage after burn, and phalloidin can alleviate the changes to a certain extent.
