摘  要:目的:激素体外定向诱导大鼠骨髓间质干细胞(MSC)分化为神经元样细胞。方法:通过贴壁法分离大鼠MSC,体外扩增培养,睾酮定向诱导MSC分化为类神经元样细胞。光镜下观察细胞形态,免疫细胞化学检测神经细胞特异性抗原标志。结果:大鼠骨髓间质干细胞可通过贴壁法成功分离并可在体外大量扩增。睾酮诱导6h后大部分MSC转变为神经元样细胞,出现胞体和突起,免疫细胞化学染色NSE、Nestin呈阳性、GFAP阴性。结论:大鼠骨髓间质干细胞可在体外经睾酮诱导分化为神经元样细胞。Objective :To investigate the effect of testosterone in in-vitro oriented induction of the differentiation of rat mesenchymal stem cells into neuron-like cells. Methods: Mesenchymal stem cells were separated from bone marrow of rats by wall sticking method, amplifying cultured in vitro, and differentiated into neuron-like cells by oriented induction with testosterone. The morphology of cells was observed under light microscopy,. Neuro-specific enolase (NSE), nestin and glial fibrillary acidic protein (GFAP) were detected by immunocytochemistry. Results: Rat mesenchymal stem cells could be separated and amplified in vitro. After being induced by testosterone for 6 hrs, most of the cells would be differentiated into neuron-like cells. By immunochemical staining, cells showed positive of NSE, nestin, and negative of GFAP. Conclusion: Rat mesenchymal stem cells could be induced to differentiate into neuronlike cells with testosterone.
