摘 要:目的 研究测定人血浆中的氟尿嘧啶浓度的色谱方法。方法 色谱柱为C_(18)柱(大连依力特,250nm×4.6mm×5μm),柱温25℃,流动相为KH_2PO_4-H_3PO_4缓冲液(0.01M,pH2.95),波长269nm。结果 回归方程为Y=2.2368X+2.7756(r=0.9998),其线性范围为:5~2500μg/L,最低检测浓度为2μg/L(S/N=3)。结论 本方法简单、快速、准确,适用于临床氟尿嘧啶的浓度测定及治疗药物监测。Objective To develop a RP-HPLC method for determination of 5-fluorouracil (5-Fu) in human plasma. Methods C_(18) column(Elite, 250 nm×4.6mm×5μm) with temperature at 25℃ was used, the mobile phase was KH_2 PO_4-H_3PO_4 (0. 01M, pH2. 95) and the UV wavelength was 269 nm, the injection volume was 50 μL. Results The 5-Fu linear equation was Y=2.2368× +2.7756 (r=0.9998) with the range from 5 to 2500μg/L(S/N=3). Conclusion This method appeared to be simple, rapid and accurate. It is suitable for the clinical determination and therapeutic drug rronitoring (TDM) of drug concentrations of 5-Fu in human plasma.
