摘  要:目的 研究蒿属花粉过敏体质者与非过敏对照者扁桃体淋巴细胞在特异性抗原刺激下细胞因子的动态变化以及细胞增殖情况。方法 采用淋巴细胞分离液分离扁桃体淋巴细胞 ,16 4 0培养液进行培养 ;经蒿属花粉抗原刺激后 ,酶联免疫吸附试验 (ELISA)检测上清液中IL 4、IL 5、IFN γ水平 ,MTT测定淋巴细胞增殖情况。结果 蒿属花粉过敏体质者扁桃体淋巴细胞经特异性抗原刺激后IL 4、IL 5水平明显升高 ,而IFN γ水平明显降低 ,与非过敏对照者细胞因子水平相比较有显著差异 (P <0 .0 1)。此外 ,过敏体质者淋巴细胞在抗原刺激后细胞增殖明显 (P <0 .0 1) ,而对照组无显著性变化 (P >0 .0 5 )。结论 过敏体质者扁桃体的淋巴细胞在蒿属花粉特异性抗原刺激下 ,Th2型细胞因子分泌升高 ,而Th1型细胞因子分泌下降 ,Th1向Th2漂移。Objective To investigate the proliferation and the dynamic changes of cytokines in tonsil lymphocytes in Artemisia pollen-atopic people and nonatopic people after stimulated by the specific antigen.Methods Tonsil lymphocytes were collected by lymphocytes separation medium and incubated with Artemisia pollen antigen.The levels of IL-4,IL-5,IFN-γ were measured by enzyme linked immunosorbent assay(ELISA).The proliferation of lymphocytes were detected with MTT method.Results The levels of IL-4,IL-5 in tonsil lymphocytes in Artemisia pollen-atopic people after stimulated by specific antigen were higher than nonatopic controls,but the levels of IFN-γ were lower than those in nonatopic controls(P<0.01,respectively).Obvious proliferation were observed in atopic group after stimulated by the specific antigen(P<0.01),but there were no significant changes in control group.Conclusion The cytokines of Th cell in tonsil lymphocytes from Artemisia pollen-atopic people after specifically stimulated by Artemisia pollen antigen lost the balance,indicating that Th1 drifts to Th2.
