三羟异黄酮对去势雌性大鼠主动脉内皮依赖性超极化的影响

摘　　要：目的　观察三羟异黄酮对去势大鼠主动脉内皮依赖性超极化的影响。方法　 4 0只成年雌性大鼠随机分成 4组 :假手术组、去势组、去势雌二醇组、去势GST组。去势 1周后开始给药 ,6周后处死大鼠。放免法测定血中雌二醇水平 ,常规微电极技术观察主动脉平滑肌细胞的内皮依赖性超极化反应。结果　去势大鼠主动脉内皮依赖性超极化较假手术组减弱 (P <0 .0 5 ) ,补充雌二醇和三羟异黄酮后 ,内皮依赖性超极化作用明显改善。结论　三羟异黄酮可加强去势雌性大鼠主动脉内皮依赖性超极化 ,这可能是雌激素和植物雌激素对绝经后妇女心血管保护作用的机制之一。Objective To investigate the effect of genistein on the endothelium-dependent hyperpolarization in aorta of the ovariectomized rats.Methods Forty female Wistar rats were randomly divided into four groups:group A:sham operation;group B:ovariectomy;group C:ovariectomy with estrogen replacement therapy;group D:ovariectomy with genistein injection.Six weeks after giving medicine,the rats were killed and conventional microelectrode technique was used to measure the Ach-induced endothelium-dependent hyperpolarization in smooth muscle cells of the aorta.Results The endothelium-dependent hyperpolarization decreased significantly in group B when compared with group A,C and D(P<0.05) and improved after giving estradiol and genistein.Conclusion The results demonstrated that genistein can enhance the endothelium-dependent hyperpolarization in aorta of ovariectomized rats and this may be the mechanism of cardiovascular protection for estrogen and phytoestrogen in menopause women
