摘  要:目的 探讨大血管缺氧对Wistar大鼠一氧化氮 (NO)活性的影响。方法 制备Wistar大鼠胸主动脉缺氧模型 ,放射免疫检测NO活性指标cGMP含量的变化。结果 内皮细胞完整 :与对照组比较 ,缺氧组主动脉cGMP降低 80 .2 8% (P <0 .0 1) ;与缺氧组比较 ,L Arg灌流、L NNA灌流组和NTG灌流组主动脉cGMP分别增加2 78.5 7% (P <0 .0 1)、降低 4 2 .86 % (P <0 .0 1)和增加 914 .2 9% (P <0 .0 1) ;与NTG组比较 ,MB +NTG组主动脉cGMP降低 35 .94 % (P <0 .0 1)。去内皮 :缺氧组对NO的影响与内皮完整相似 ;L Arg组、L NNA组均不明显改变血管cGMP水平 ;NTG组cGMP水平明显增加 ,NTG +MB组可部分阻断该作用。结论 大血管缺氧显著抑制NO生成与释放 ,造成NO生成缺陷和活性降低 。Objective To observe effect of large blood vessel hypoxia on the Wister rat's NO activity.Methods Thoracic aorta anoxia model of Wistar rat was prepared and concentration change of cGMP was examined by radioimmunoassay,a NO active index.Results In endothelium complete condition,hypoxia group vs control group,cGMP concentration reduces 80.28%(P<0.01);with hypoxia group comparison,cGMP level of L-Arg group,L NNA group and NTG group respectively increased 278.57% (P<0.01),reduced 42.86% (P<0.01) and increased 914.29% (P<0.01);compared with NTG group,MB+NTG group caused cGMP reduce 35.94% (P<0.01).In endothelium taken off condition,Hypoxia group was similar to endothelium complete condition for the influence of NO.However L-Arg group or L NNA group did not change the cGMP level of blood vessel obviously;NTG group increased cGMP level obviously,and it was partially inhibited by NTG+MB group.Conclusion These results suggest that large blood vessel anoxia markedly inhibit NO generation and release,and make NO generation deficiency and decrease its activity.It causes that endothelium dependent blood vessel relaxation is abnormity reduced.
