摘  要:通过细胞遗传学方法对转基因鲤鲫杂交 (F1)回交三倍体子代鱼进行了红细胞及其核大小的测量和DNA含量的测定 ,结果为 :采用血涂片法测量鲤鲫杂交回交子代红细胞面积和体积分别是二倍体红鲫鱼的 1.4 3倍和 1.5 7倍 ,接近三倍体的理论预期值 1.5倍 ;通过流式细胞仪对鲤鲫杂交子代细胞和二倍体细胞进行相对DNA含量测定 ,结果为 :在所测的 12尾鱼中 ,有 9尾鲤鲫杂交回交子代鱼的DNA含量是对照鱼的 1.5倍 ,占总鱼数的75 % ,在 2~ 3倍体之间非常接近三倍体 (非整倍三倍体 )的鱼有 3尾 ,占总鱼数的 2 5 % ,本结果与其细胞染色体核型分析结果基本一致。A study was conducted to determine red blood cell (RBC) and its nucleus size as well as DNA content in backcross progenies of hybrids produced by transgenic common carp and transgenic crucian carp by using cytogenetic methods. DNA content of backcross progenies and diploids were measured by the flow cytometry. The results show that the area and volume of RBC in backcross progenies are very close to 1.5 times as much as the theoretical expecting value of the triploids by using blood-painting method, and they are 1.43 and 1.57 times as large as those of RBC in diploids respectively. Among 12 measured progenies, the DNA contents in 9 progenies are 1.5 times as much as that of the control group, accounting for 75% of the total progenies. 3 progenies are very near to triploid (refer to as non-integral triploid), accounting for 25% of the total ones. To sum up, the results obtained are consistent with those of karyotypic analysis of cell chromosome.
