益气活血中药对血管紧张素Ⅱ致培养乳鼠心肌细胞肥大的影响

摘　　要：目的　通过研究益气活血药对血管紧张素Ⅱ (AngⅡ )致心肌肥大的影响 ,寻找能抑制血管紧张素Ⅱ所致心肌肥大的益气活血中药。方法　通过在培养乳鼠心肌细胞加入血管紧张素Ⅱ ,观察细胞总蛋白质、细胞直径和细胞数 ;在此基础上 ,进行益气活血中药对血管紧张素Ⅱ致培养乳鼠心肌细胞肥大实验药理学的研究。选用药物中 ,采用其有效成分丹参素、川芎嗪以及黄芪注射液 ,而党参和党参加丹参则采用血清药理学方法。结果　AngⅡ加Losartan组和AngⅡ加丹参大、中、小剂量组分别减少心肌细胞总蛋白质含量14 .0 % (P <0 .0 5 )、10 .9% (P <0 .0 5 )、7.7%和 6.4% ,减少心肌细胞直径 2 2 .0 % (P <0 .0 1)、16.0 % (P <0 .0 5 )、13 .9% (P <0 .0 5 )和 3 .4% ,而对心肌细胞数量无明显的影响 ;AngⅡ加川芎嗪大、中、小剂量组分别减少心肌细胞总蛋白质含量 11.6% (P <0 .0 5 )、6.8%和 4.0 % ,减少心肌细胞直径16.0 % (P <0 .0 5 )、9.1%和 66% ,各组心肌细胞数量无明显差异 ,表明丹参素和川芎嗪与Losartan一样可以减少血管紧张素Ⅱ所引起的心肌细胞蛋白质含量的增加以及直径的增加 ,并呈量效关系 ,而对心肌细胞数量无明显的影响 ,不引起心肌细胞增殖 ;而益气药党参、黄芪和党参对丹参组无此作用。Objective The objectives are to discover some Qi tonic and drugs for promoting blood circulation of Chinese herbs who can prevent the myocardial hypertrophy induced by AngⅡ.Methods The studies were also done to understand the effects of Qi tonic and drugs for promoting blood cirulation of Chinese herbs on the myocardial hypertrophy induced by angiotensin Ⅱ in the cultured neonatal myocardial cells in vitro.The efficient components of Danshensu,Ligustraxine,and Astrgalus Root injection were elected,while serum pharmacology of Dangshen and Dangshen plus Danshensu were used.Results It showed that Danshensu and Ligustrazine reduced the increase of content of the total cellular protein and cellular diameter induced by nagiotensinⅡin the cultured neonatal myocardial cells in vitro,so it demonstrated that Danshensu and Ligustrazine were same as the Losartan who reduced the total cellular protein of myocardial cells and the cellular diameter and also had dose-effect relationship on these effects;there were no significant differences on the number of the cells for their groups,so it demonstrated myocardial cells did not proliferate in the cultured neonatal myocardial cells in vitro.While the Dangshen,Astrgalus Root and Dangshen plus Danshen had no such effects.Conclusion It showed that the drugs for promoting blood circulation can inhibit hypertrophy of cardiac myocytes induced by angiotensinⅡin the cultured neonatal myocardial cells in vitro.The blood stasis is the basis of LVH and promoting blood circulation may prevent LVH.The researches provide experiment basis for further clinical study.
