贯叶连翘薄膜片中金丝桃素和金丝桃素类化合物的HPLC和UV测定

摘　　要：建立了RP-HPLC法和UV法分别测定贯叶连翘薄膜片中金丝桃素和总金丝桃素类的含量。RP-HPLC法采用ODS柱,流动相为5%甲醇(含0.1%磷酸,三乙胺调至pH 4.5)-甲醇-乙腈(15∶40∶45),检测波长588nm。UV法采用甲醇避光超声提取制备样品溶液,588 nm处测定。金丝桃素和总金丝桃素类的线性范围分别为0.04～0.24mg和2～12mg/ml。平均回收率分别为103.8%和103.3%。HPLC and UV methods for determination of hypericin and hypericins respectively in Hypericumperforatum film-coated tablets were established. For HPLC method, an ODS column was used with the mobile phase of5% methanol (containing 0.1% phosphoric acid, adjusted pH to 4.5 with triethylamine)-methanol-acetonitrile (15∶40∶45). For UV method, light-proof ultrasonic extraction was used to prepare the sample solution with methanol assolvent, at the detection wavelength of 588 nm. The calibration curves were linear in the ranges of 0.04～0.24mg(hypericin)and 2～12mg/ml(hypericins), respectively. The recoveries were 103.8% and 103.3%, respectively.
