摘  要:目的 :观察糖尿病大鼠视网膜组织中细胞凋亡量的变化。方法 :4 0只 Wistar大鼠被随机分成实验组和对照组 ,每组 2 0只。实验组用链脲佐菌素诱发糖尿病 ,在糖尿病发病后 4、6、1 2和 1 6周检测视网膜细胞凋亡和视网膜组织中钙镁离子浓度的变化 ,并与空腹血糖、糖化血红蛋白及糖尿病病程的关系进行分析。结果 :糖尿病大鼠视网膜细胞溶解液中在发病后 4周可测到细胞凋亡 ,且随糖尿病病程的延长凋亡程度逐渐加重 ;同时伴有视网膜组织内钙离子浓度的增加、镁离子浓度减少。双变量相关分析表明 ,糖尿病大鼠视网膜细胞凋亡的程度与空腹血糖浓度、糖化血红蛋白水平、视网膜内钙镁离子含量及糖尿病病程密切相关。结论 :糖尿病大鼠视网膜中存在细胞凋亡和离子含量的异常 。Objective:To observe the changing of cells apoptosis quantity in retina of diabetic rats.Methods:40 wistar rats were divided into two groups randomly: experimental group and control group. The rats in experimental group were induced to develop diabetes by streptocotocin. The apoptosis cells quantity in retinal cells of diabetic rats were detected by ELISA, and the content of Ca 2+ , Mg 2+ in retina tissue were measured by atomic absorption spectrophotometry after 4,6,12 and 16 wk. The relation of the apoptosis quantity with levels of fasting blood glucose (FBG), hemoglobin glycosylation (HbA 1 c), the content of Ca 2+ and Mg 2+ in retinal tissue, and the course of disease was also analyzed.Results:The cells apoptosis in the retinal tissue of diabetic rats was detected in 4wk after diabetes. With the development of diabetic course, the extent of cells apoptosis in retinal tissue was more serious. Content of Mg 2+ in retinal tissue decreased whereas Ca 2+ increased. The changes of cells apoptosis connected with the changes of FBG, HbA 1 c, the content of Ca 2+ and Mg 2+ in retinal tissue, and the course of disease.Conclusion:There are cells apoptosis and abnormal of ion content in the retinal tissue of rats with diabetic mellitus. The changes may be one of the pathologic basis of diabetic retinopathy.
