摘 要:目的比较4种聚丙烯酰胺凝胶银染方法,为初次使用者选择方法时提供参考。方法用24 mer的引物作为上样样本,分别采用氨水法、低浓度硝酸银法、0.1%及0.2%硝酸银法染色。后2种配合不同的显色剂显色。结果前2种未显色,0.1%及0.2%硝酸银可显色。结论0.2%的硝酸银与氢氧化钠显色剂相结合具有最高的灵敏度;0.2%的硝酸银与碳酸钠显色剂相结合具有最佳的显像效果。Objective To compare 4 silver staining methods for DNA detection in polyacrylamide gel electrophoresis. Methods After the electrophoresis was completed, the gels were stained separately by four different methods, namely using ammonia-silver-citric acid, low-concentration silver nitrate, 0.1% and 0.2% silver nitrate. Results DNA was detected only by staining with 0.1% and 0.2% silver nitrate. Conclusion 0.2% silver nitrate used along with sodium hydroxide shows the highest sensitivity in DNA detection, while its use with sodium carbonate produces the best quality of the image.
