摘  要:目的 观察神经干细胞 (neuralstemcell,NSC)增殖分化相关蛋白在左旋单钠谷氨酸 (monosodiumglutamate ,MSG)大鼠NSC增殖分化中的作用。方法 提取前脑组织蛋白、利用Westernblot方法检测NSC增殖分化相关蛋白变化。结果 与正常对照组比较 ,造模后 3 0、60d、MSG大鼠NSC增殖分化相关蛋白Notch1、hes5、Mash1、NeuroD表达水平显著降低 ,尤以Notch1和hes5更显著 (P <0 0 1) ;与实验组比较 ,左归丸可明显增加Notch1、hes5、Mash1、NeuroD表达水平 (P <0 0 1)。结论 MSG显著抑制NSC增殖分化相关蛋白表达水平 ,左归丸能部份拮抗MSG对NSC增殖分化相关蛋白表达的抑制。Objective To investigate the roles of differentiation and proliferation related proteins in neural stem cells (NSCs) in rats treated with L monosodium glutamate (MSG). Methods Tissue proteins were extracted from the rat forebrain. Western blotting was used to investigate the changes of differentiation and proliferation related proteins in NSCs in rats treated with MSG. Results Compared with those in rats of the normal control group, the related proteins (Notch1, hes5, Mash1, and NeuroD) in NSCs were significantly decreased in MSG rats at 30 and 60 d, especially for Notch1 and hes5 ( P <0.01). Compared with those in the model group, the levels of Notch1, hes5, Mash1, and NeuroD were significantly increased in MSG rats after treatment with the Zuogui pills at 30 and 60 d ( P <0.01). Conclusion MSG could effectively decrease the levels of Notch1, hes5, Mash1, and NeuroD which are related with the differentiation and proliferation of NSCs. Zuogui pills could partly reverse the inhibition of MSG. Notch family and bHLH transcription factor are probably involved in the NSC proliferation and differentiation induced by Zuogui pills.
