Objective To study telomerase activity in mononuclear cells (MNCs) of bone marrow (BM) specimens from pediatric patients with acute myeloid leukemia(AML) at diagnosis and after chemotherapy. For comparative analysis,MNCs from control donors were analyzed. Methods Telomerase in 29 cases of children with AML were evaluated using a modified telomeric repeat amplification protocol (TRAP) assay. Results At diagnosis, high telomerase activity was detected in MNCs from AML children (35 55±37 41TPG).This exceeded by 15 fold the activity in MNCs from control donors(2 36±2 27TPG,n=9) (P<0 01).After chemotherapy, telomerase activity in MNCs from the AML children decreased to (2 66±3 99) TPG,and 10 cases of the AML children less than 1 5TPG.Conclusion Telomerase is highly upregulated in BM but decreased after induction therapy;and it is correlated with remission in AML children. Our findings suggest that telomerase upregulating plays an important role in the development of AML and may be a useful predictor for prognosis in childhood leukemia.
