摘  要:用改良的McCoy's5a培养液(含3mML-谷氨酰胺、0.1%BSA、29ng/mL睾酮、2.5μg/mL转铁蛋白、100ng/mL胰岛素和4ng/mL硒),在96孔培养板中,每孔250μl培养液的条件下,研究了神经生长因子(NGF)对牛腔前卵泡体外发育的影响。结果表明:在培养液中添加三种不同浓度NGF(4、6、8IU/ml)对体外培养的Φ<60μm腔前卵泡有促进生长的作用。在体外培养10d时,三种不同浓度NGF组的Φ<60μm腔前卵泡及其卵母细胞直径平均增长值分别为27.84±4.51μm和6.11±1.81μm,显著高于对照组(分别为8.09±2.66μm和0.26±0.06μm)(p<0.01)。但对60μm>Φ>130μm和Φ>130μm的腔前卵泡作用不显著(p>0.05)。同时,不同浓度的NFG显著提高了Φ<60μm腔前卵泡的发育率,对照组为11.76%(2/17),而三个实验组平均为17.65%(7/40)(p<0.05)。A serum-free culture system using McCoy's 5a culture medium supplemented with 20mM Hepes, 3mM L-glutamine, 0.1% BSA, 29ng/mL testosterone, 2.5μg/mL transferring, 100ng/mL insulin and 4ng/mL selenium for culture of bovine preantral follicles were developed Under conditions of 250μl McCoy's 5a culture medium in each well of 96-well cuture plates and one follicle in each well, the effects of NGF on in Vitro Development of Preantral Follicles were studied, The results showed that different concentrations of NGF (4-8IU/ml) in the culture medium promoted the in vitro development and growth of preantral follicles (Φ<60μm), Increment of diameter of preantral follicles and its oocyte in experimental group was higher than in contral group (average increment of diameter of preantral follicles its oocyte in experimental group were 27.84±4.51μm and 6.11±1.81μm respectively, and that of in contral group were 8.09±2.66μm and 0.26±0.06μm) respectively (p<0.01). in the meantime different concentrations of NGF (4-8IU/ml) in the culture medium promoted rate of upgrowth [average rate of upgrowth of preantral follicles in experimental group was 17.65%(7/40), and that of in contral group was 11.76%(2/17)] (p<0.01). however, those functions of NGF on preantral follicles (60μm>Φ>130μm and Φ>130μm) were not shown obviously (p>0.05).
