摘 要:目的 :建立高效液相色谱法分离并测定洁康舒洗剂 (大黄、黄柏、苦参等 )中大黄酸、大黄素及大黄酚的含量。方法 :采用SymmetryC18色谱柱 (3.9mm× 15 0mm ,5 μm) ,SymmetryC18预柱 (5 μm) ,流动相为甲醇 水 磷酸 (82∶18∶0 .1) ,柱温2 4℃ ,检测波长 4 32nm ,流速 1.0mL·min-1。结果 :本法测定大黄酸、大黄素及大黄酚的线性范围分别为 :7.6 0~ 38.0 0 μg·mL-1(r=0 .9994 ) ,1.75~ 8.75 μg·mL-1,(r =0 .9999) ,2 .4 0~ 12 .0 0 μg·mL-1(r =0 .9999) ;平均回收率 (n =5 ) :大黄酸为 98.0 2 % ,RSD =0 .39% ,大黄素为 96 .6 3% ,RSD =0 .71% ,大黄酚为 97.0 5 % ,RSD =0 .5 1%。结论 :方法简便、结果准确、重现性好 。AIM: To establish HPLC method for separation and determination of rhein, emodin and crysophanol in Jiekangshu lotions(Radix et Rhizoma Rhei, Cortex Phellodendri, Radix Sophorae Flavescentis,etc.). METHODS: The chromatographic column: Symmetry C 18 column(3.9mm×150mm,5μm) and in advance column: Symmetry C 18 (5μm) were used with the mobile phase of methanol-water-phosphate acid(80∶20∶0.1), 1.0mL·min -1, flow rate, column temperature at 24℃ and detection wavelength at 432nm. RESULTS: Rhein, emodin and chrysophanol in Jiekangshu Lotion showed the good linear relationships at the range of 7.60~ 38.00μg·mL -1 (r= 0.9994), 1.75~8.75μg·mL -1 (r=0.9999), 2.40~12.02μg·mL -1 (r=0.9999), respectively. Their average recoveries: Rhein was 98.02%, Emodin 96.63% and Chrysophanol 97.05%, respectively. CONCLUSION: This method is simple. The results are accurate and reproducible.It can be used in the quality control of Jiekangshu lotion.
