摘 要:目的 观察汉坦病毒 (HV)对人体骨髓细胞的作用。方法 应用免疫荧光技术、光镜和电镜细胞学检查对骨髓细胞结构尤其是超微结构进行观察研究 ,并设对照组。结果 检测了 2 7例肾综合征出血热 (HFRS)患者骨髓细胞的病毒抗原 ,2 0例呈现阳性。发现粒细胞含HV抗原阳性细胞百分数最高 (76 % ) ,其他依次为单核细胞 (6 5 % ) ,淋巴细胞 (40 % ) ,巨核细胞 (2 0 % )和红细胞 (3 7% )。利用光镜、透射电镜和扫描电镜进行细胞学观察发现感染病毒的骨髓细胞、细胞膜和细胞器的损伤明显高于对照组。结论 HV可侵入骨髓细胞中 ,并对人骨髓细胞具有致病变作用。Objective To observe cytopathogenic effect of Hantaan virus (HV) on cultured human bone marrow cells. Methods Light and transmission electron microscopy and direct immunofluorescent technique were applied to study cellular structrue especially ultrastructural changes of bone marrow cells from patients with Hantaan virus infection. Bone marrow cells of one healthy volunteer were also studied as control. Results The antigen of HV was found in bone marrow cells of 20 of 27 HFRS patients by the aid of direct immunofluorescent technique. It was found that the granulocytes had the highest percentage of HV antigen positive cells(76%),followed by monocytes(65%),lymphocytes(40%),megakaryocytes(20%) and the lowest was found in erythrocytes (3.7%). The injury of cell membrane after infection with HV was significantly more severe than that in the control group under the light and electron microscopy. Conclusion This study demonstrated that HV could attack humman bone marrow cells and cause cytopathogenic effect on them.
