调肝导浊中药对体外培养大鼠主动脉平滑肌细胞增殖调控的影响

摘　　要：[目的]观察调肝导浊中药对大鼠主动脉平滑肌细胞 (SMC)内Ca2 +浓度、C_myc表达和SMC增殖的影响。[方法]采用体外SMC技术 ,免疫组织化学方法测量SMC的增殖核抗原(PCNA)阳性细胞率和C_myc阳性表达率 ,通过负荷钙离子荧光指示剂Fura_2/AM进行[Ca2 +]i 测定。[结果]高脂血清培养组SMC的PCNA阳性细胞率和C_myc阳性表达率明显高于正常培养(P<0.01)。调肝导浊中药通过降低胞内[Ca2 + ]i,抑制C_myc表达 ,使该组的PCNA阳性率明显低于高脂血清组(P<0.01)。[结论]调肝导浊中药对SMC增殖起到良好的调控作用 ,从而有效地防止动脉粥样硬化 (AS)的发生与发展。Objective] To observe the influence of“Tiaogandaozhuo”herbs on concentration of Ca 2+ ,expression of C-myc and proliferation of SMC.[Methods]PCNA positive cell rate and C-myc positive expression rate were detected by external SMC technique,and immunohistochemical method,[Ca 2+ ] was determined by load fluorescence indicator Fura-2/AM.[Result] The PCNA positive cell rate and C-myc positive expression rate in high-fat serum culture group was higher than that of normal group(P<0.01).“Tiaogandaozhuo”herbs made the PCNA positive rate obviously lower than that of control group by descending [Ca 2+ ]and inhibiting of the expression of C-myc,(P<0.01).[Conclusion]“Tiaogandaozhuo”herbs possess good regulating and controlling effect on the proliferation of SMC,and hence effectively prevent the occurring and development of atherosclerosis(AS).
