抗炎灵汤剂对MODS大鼠iNOS、IL-6和TNF-α表达的影响

摘　　要：目的 :探讨抗炎灵汤剂对MODS大鼠结肠IL - 6、TNF -α和iNOS的作用。 方法 :将Wistar大鼠分成两组 :MODS模型组和中药抗炎灵防治组 ,分别观测单位时间排便的粪点数、血清NO的变化 ,取远端结肠平滑肌分别行病理学、免疫组化和RT -PCR方法检查。 结果 :防治组与模型组相比排便点数明显增加 ;而防治组血清NO含量、结肠平滑肌炎性反应、IL - 6、TNF -α和iNOS以及IL - 6mRNA、TNF -αmRNA和iNOSmRNA的表达较模型组降低 (P <0 0 5 )。 结论 :抗炎灵汤剂可改善MODS大鼠结肠平滑肌炎性细胞浸润 ,减少MODS大鼠结肠iNOS、IL - 6和TNF -α的生成 ,促进结肠运动。Objective To explore the effect of Kang YanLing on interleukin-6(IL-6),tumor necrotixing factor-α(TNF-α and induced nitric oxide synthase(iNOS) in the colon of the rats after multi-organ dysfunction syndrome(MODS). Methods Twenty Wistar rats of MODS models were divided into two groups,the control group and the Kan YanLing treatment group.The mumber of stool and serum nitric oxide(NO)level in the two groups were measured.The distal colon amooth muscle were removed and observed pathologically and immunohistochemically determined and by retrotransmission polymerase chain reaction methods.Results The number of stool in the treatment group were higher than in the MODS control;the concentration of serum NO,the inflammatory response in the colon,the expressions of IL-6,TNF-α and iNOS and the expression of IL-6mRNA,TNF-α MRNA and iNOSmRNA in the treatment group were lower than in control(P<0 05). Conclusion Kang YanLing could reduce the infiltration of polymophonuclear meutrophilic granulocytes into the muscular layer,decrease the xpression of iNOS,IL-6 and TNF-α and promote colon motility in MODS rats.
