摘  要:目的 :探讨孕早期干预协同刺激信号对自然流产模型孕鼠免疫活性细胞MHC Ⅱ类抗原和协同刺激分子的调控作用。方法 :建立自然流产模型组CBA×DBA 2和协同刺激信号干预组CBA×DBA 2 (CBA小鼠在孕 4天腹腔注射CD80及CD86mAb各 0 1mg)。分别于孕第 14天计数两组的胚胎吸收率 ,于孕第 9天采用流式细胞技术分析孕鼠脾细胞MHC Ⅱ类抗原与协同刺激分子CD80 (B7 1)、CD86 (B7 2 )、CD2 8和CD15 2 (CTLA 4 )的表达。结果 :孕早期干预协同刺激信号后 ,孕第 14天的胚胎吸收率显著下降 (P <0 0 5 ) ;孕第 9天免疫活性细胞CD80、CD86和CD2 8的表达显著下降 (P <0 0 1) ;CTLA 4的表达则显著升高 (P <0 0 5 ) ;而MHC Ⅱ类抗原的表达无显著改变 (P >0 0 5 )。结论 :孕早期体内干预协同刺激信号使免疫活性细胞协同刺激分子CD80、CD86、CD2 8表达下调和CTLA 4表达上调 ,及母胎界面Th2型免疫偏离 ,从而诱导系统性母Objective:To explore the effect of blocking the costimulatory signal at the early gestation on expression of costimulatory molecules and MHC-Ⅱ class antigen of immuno-competent cells in the abortion-prone model.Methods:After the abortion-prone CBA×DBA/2 matings were established,the pregnant CBA females were intraperitoeally injected with purified rat anti-mouse CD86/CD80mAb 0.1 mg at day 4 of gestation.Expressions of MHC-Ⅱ antigen and costimulatory molecules on the immuno-competent cells of spleen were analyzed by flow cytometry(FCM)at day 9 of gestation respectively.The embryo resorbing rate was counted at day 14 of gestation.Results:After in vivo blocking the costimulatory signal in the abortion-prone model,the embryo resorbing rate was reduced significantly(P<0.05),the expression of costimulatory molecules CD80,CD86 and CD28 on the splenic immuno-competent cells decreased significantly(P<0.01),while that of CTLA-4 increased significantly(P<0.05),that of MHC-Ⅱ antigen was invariable.Conclusion:The systemic materno-fetal immune tolerance induced by blocking the costimulatory signal may be attributed to the down-regulated expression of costimulatory molecules(CD80、CD86 and CD28)and up-regulated expression of CTLA-4 on the immuno-competent cells except for Th2 bias at the materno-fetal interface.
