摘 要:目的:探讨不同剂量氟化钠 (Na F)对兔体外破骨细胞功能的影响。方法:从新生兔四肢长骨中分离破骨细胞 ,采用体外培养破骨细胞的方法 ,用倒置相差显微镜和计算机图像分析技术 ,测量在含有不同浓度 (1.0 0×10 - 7~ 1.0 0× 10 - 4m ol/ L )的 Na F培养液中骨吸收陷窝数和表面积。结果:染氟 1.0 0× 10 - 6 、1.0 0× 10 - 5和 1.0 0×10 - 4m ol/ L 组骨片上骨吸收陷窝数量和表面积与对照组比较均减少 (P <0 .0 1) ,而且骨吸收陷窝数及表面积随氟浓度的增加而减少。 结论 :氟对体外培养的破骨细胞有直接的损伤作用。Objective: In order to explore the effects of different concentrations of NaF on resorptive activity of isolated osteoclast. Methods: Osteoclast was obtained from the 24-h newborn Japanese rabbit. The effects of NaF on the activity of isolated rabbit osteoclastic culture on thin slices of devitalized compact bovine bone were analyzed. Osteoclastic resorption was quantitated by counting the number of resorption lacunae and measuring their surface area and their depth by using scanning electron microscopy. Results: The results showed that NaF in different concentrations of NaF (1.00×10 -4 ?1.00×10 -5 和1.00×10 -6 mol/L) decreased the number of resorption lacunae made by individual osteoclasts and decreased the resorbed area per osteoclast. Conclusion: NaF could directly injury osteoclast and inhibit bone resorption function of osteoclast in vitro.
