妊高征胎盘滋养细胞浸润能力的检测及胰岛素样生长因子-II、转化生长因子-β1对其的影响

摘　　要：目的　检测妊娠高血压综合征 (妊高征 )胎盘滋养细胞浸润能力 ,胰岛素样生长因子 II(IGF II)、转化生长因子 β1(TGF β1)对细胞滋养细胞侵袭力的影响。　 方法　从正常及妊高征胎盘分离、纯化细胞滋养细胞行体外培养 ;用体外侵袭试验检测正常及妊高征胎盘滋养细胞的侵袭力 ;IGF II、TGF β1对两组滋养细胞侵袭力的影响。　 结果　妊高征滋养细胞较正常滋养细胞的侵袭力显著降低 (P <0 .0 5 ) ;IGF II明显促进、TGF β1明显抑制正常妊娠滋养细胞的侵袭能力(P <0 .0 5 ) ;二者对妊高征滋养细胞侵袭力的影响不显著 (P >0 .0 5 )。　结论　妊高征胎盘滋养细胞的侵袭力显著降低 ;IGF II、TGF β1可能在妊高征胎盘滋养细胞浅浸润过程中起一定作用。Objective: To investigate the trophoblast invasion activity of pregnancy-induced hypertension syndrome (PIH) and to evaluate the effects of IGF-II and TGF-β1 on the cytotrophoblast invasion.Methods:(1)The Cytotrophoblasts were separated and purified from the normal and PIH placenta. (2)cytotrophoblast invasion activity of normal and PIH placenta was measured by an in vitro invasion assay. (3)The effects of IGF-II and TGF-β1 on cytotrophoblast invasion were studied.Results: (1) In PIH group,the invasive ability of cytotrophoblasts was dramatically decreased. (2) In normal group,trophoblast invasion was enhanced by IGF-II and inhibited by TGF-β1 significantly. (3) The effect of IGF-II or TGF-β1 on trophoblast invasion of PIH was not statistically significant. Conclusion: These results suggested that the cytotrophoblast invasive ability of PIH was dramatically decreased,and IGF-II and TGF-β1 may have an important role in the shallow invasion of PIH trophoblast.
