摘  要:目的 :观察牛嗜铬细胞 (BCC)植入大鼠脊髓蛛网膜下腔后的镇痛效应。方法 :5 0只成年Sprague Dawley大鼠 ,随机分为A、B、C、D和E组 ,每组 10只。A、B、C和D组大鼠蛛网膜下腔植入 2 0 μlBCC悬液 ;E组大鼠植入等量的DMEM液。D组和E组于移植手术后 ,每周进行急性热刺激实验 ,持续 8周 ;A、B和C组于移植手术后 3周分别进行纳洛酮、酚妥拉明和生理盐水阻断的福尔马林实验 ,D组和E组进行福尔马林实验。结果 :急性热刺激实验中D组抗热伤害效应明显高于E组 (P <0 .0 5 ) ,并持续 8周 ;福尔马林实验中D组急性期反应评分和慢性期反应评分均低于E组 (P <0 .0 1)。阻断剂实验中 ,急性期疼痛评分A组、B组均高于C组 (P <0 .0 5或P <0 .0 1) ,但慢性期2组间差异无统计学意义 (P均 >0 .0 5 )。结论 :BCC移植对大鼠急慢性痛均有明显镇痛作用 ,植入的BCC分泌的脑啡肽类和儿茶酚胺类活性成分参与镇痛过程的调节 ,但 2种物质的作用机制不同。Aim:To evaluate the analgesic effects of bovine cromaffin cells(BCC)implanted into the rat spinal subarachnoid spole. Methods: Fifty Sprague-Dawley rats weighing 280~320 g were randomly devided into A,B,C,D,and E groups with 10 rats in each group. 20 μl BCC suspensions containing 4×10 7 cells were injected into the spinal subarachnoid of A,B,C, and D groups. The rats in group E were given the same volume of DMEM.The rats in A,B, and C groups were respectively stimulated by formalin with pretreatment of naloxone, phentolamine, and saline solution after 3 weeks of transplantation. The rats in D and E groups were treated with formalin test after 3 weeks of transplatation and acute heat-stimulating test every weeks until 2 months after transplatation . Results: The antinociceptive scores of acute heat stimulating test in group D were higher than those in group E (P<0.05). The antinociceptive effects in group D lasted 8 weeks after implantation ,then declined with the lapse of time (P<0.05). The scores of the first and the second phases of pain response of formalin test in group D were lower than those in E group(P<0.01). In addition, the acute pain was abrogated by naloxone and partially reversed by α2-adrenergic receptor antagonist phentolamine(P<0.01). Conclusion: Antinociceptive effects of BCC implanted into subarachnoid space are remarkable in both acute and chronic pain models.The enkephalin and catecholamine secreted by the BCC may play an important role in the attenuation of pain through different mechanisms.
