摘　　要：背景:明胶酶B基因在脑血管病变中表达已有报道,但在脑损伤脑水肿形成中是否起重要作用?目的:了解大鼠脑损伤后脑组织明胶酶B基因表达情况,探讨其与脑水肿发生的关系。设计:随机对照的实验研究。地点和材料:实验地点:解放军第二五二医院动物室及中心实验室,40只Wistar大鼠,雌雄各半,体质量(410±50)g。干预:40只大鼠,于伤后不同时间取水肿区脑组织及正常脑组织:提取组织总RNA:用共扩增反转录定量PCR方法测定MMP-9基因表达水平。主要观察指标:观察MMP-9大鼠脑损伤后12,24,48h表达水平,并与正常脑组织表达水平比较。结果:脑损伤水肿区脑组织MMP-9基因表达水平明显高于正常脑组织(0.66±0.14):伤后12h表达最强(2.63±0.14),24,48h表达逐渐下降犤(1.71±0.30)和(1.30±0.26)犦,但仍高于正常对照组,差异有显著性意义(t=3.43,3.90,P<0.01)。结论:明胶酶B在脑损伤脑水肿形成中可能起重要作用。BACKGROUND:There are some reports about expression of gelatinase B in cerebrovascular diseases.However,whether it plays an important role in forming cerebral edema of brain damage is still uncertain.OBJECTIVE:To explore the gene expression of gelatinase B of rats after brain damage and discuss the relationship between it and cerebral edema.DESIGN:Randomized patient controlled trial.SETTING and MATERIALS:Animal center of No.252 Military Hospital and Central Lab.40 Wistar rats of either sex with weight of(410±50) g were chosen.INTERVENTIONS:Tissues of edema area and normal brain of 40 rats were collected in different time after damage:total RNA were abstracted and MMP-9 gene expression was tested by PCR means. MAIN OUTCOME MEASURES:Expressive level of MMP-9 of rats in the 12th,24th and 48th hours after brain damage and compare with expression in normal brain tissue.RESULTS:The expressive level of MMP-9 gene was much higher in edema area than that in normal brain tissue(0.66±0.14):the expression was the highest in the 12th hour after damage(2.63±0.14),it decreased in the 24th hour(1.71±0.30) and 48th hour(1.30±0.26) but still was higher than that of normal group.There was significant difference between two groups(t=3.43,3.90,P< 0.01).CONCLUSION:Gelatinase B may play an important role on the occurrence of cerebral edema.
