皮肤细胞悬液移植的基础研究:人黑素细胞膜片体外附着生长观察

摘　　要：目的:观察黑素细胞与BioFOLIE膜的亲和性,体外建立黑素细胞膜片,为改进细胞移植治疗白癜风做好准备。方法:把黑素细胞接种于BioFOLIE膜上,观察细胞生长形态,扫描和投射电镜观察黑素细胞和BioFOLIE膜的贴附状况。把接种黑素细胞的BioFOLIE膜按常规冻存到液氮中,复苏后观察黑素细胞的生长。结果:黑素细胞接种于BioFOLIE膜后很快贴壁。黑素细胞伸展充分,和六孔板中培养的黑素细胞形态类似。扫描电镜发现黑素细胞贴附很好,有长的树突形成。透射电镜可见细胞BioFOLIE膜粘贴紧密,膜表面形成小的裂隙,裂隙中有黑素颗粒。黑素细胞以膜片形式冻存复苏后大部分细胞存活,仍能培养传代。结论:黑素细胞在BioFOLIE膜上贴附快,伸展好,分泌黑素颗粒功能正常。两者亲和性好。用BioFOLIE膜制备的黑素细胞膜片观察容易,并能够冻存备用,使用方便。AIM:To observe the affinity of melanocytes and BioFOLIE membrane and establish the melanocyte graft in order to make preparation for improving cellular transplantation method to treat vitiligo. METHODS:The human melanocytes were bred on BioFOLIE membrane.The form of growth were observed and the affinity of melanocytes and BioFOLIE membrane was observed by scan and electron microscope.The BioFOLIE membrane that was bred in melanocyte was frozen in liguid nitrogen.The growth of melanocyte was observed after anabiosis. RESULTS:The melanocytes sticked on BioFOLIE membrane quickly and extended its dendrites,which was similar to the form of melanocytes cultivated in six pore plate.The scanning electron microscope showed that the melanocytes were well attached with the formation of long dendrite.The transmission electron microscope showed the BioFOLIE membrane was pasted tightly, and there was little crevice formed on the surface of the membrane and there was melanin granule in crevice.The melanocyte graft cauld keep good livingness after frozen,which could be cultivated and transferred. CONCLUSION:The melanocytes grow very well on BioFOLIE membrane and form melanocyte graft.The function of the excreted melanin granule is normal.The affinity of the two is good.The melanocyte graft made by BioFOLIE membrane is easy to be observed,which can be frozen for stahdby and easy to use.
