摘  要:目的:对胚胎大鼠脊髓源性神经干细胞进行分离和培养,证实其增殖潜能。方法:从孕17d的SD大鼠胚胎脊髓中分离、培养神经干细胞并用血清诱导其分化,通过免疫荧光化学方法研究其特性。结果:获得了大量未分化、呈悬浮生长的神经干细胞球,传数代后仍有干细胞特性,在血清诱导下,可分化为神经元及神经胶质细胞。结论:本实验成功分离胚胎大鼠脊髓源性神经干细胞,为神经干细胞的基础研究奠定一定基础。AIM:To isolate and culture the neural stem cells (NSCs)from fetal rat spinal cord, and to confirm the potential of proliferation. METHODS:NSCs, separated from the spinal cord of 17 day old rat fetus, were cultured and induced to differentiation in vitro,and the characteristics were studied by using histochemistry and immunofluorescent. RESULTS: A great amount of NSCs,which were undifferentiated and grew in suspension,were obtained,and they still had the characteristics of stem sells,which could differentiate into neurons, astrocytes and oligodendrocytes under the induction of serum. CONCLUSION: Spinal cord derived NSCs are successfully isolated from fetal rat, and the NSC model can be used for further basic research.
