摘  要:目的:观察肢体负压对周围动脉闭塞性病变犬背根神经节中P物质免疫反应阳性神经纤维的影响。方法:犬17只,随机分治疗组10只,非治疗组5只和正常对照组2只,治疗组和非治疗组均将动物制作左后肢缺血模型,治疗组在模型制作后14d,开始行患肢负压治疗10d(15min/次);非治疗组不做负压治疗。3组均行背根神经节免疫组化染色,检测P物质免疫反应阳性纤维。结果:背根神经节中的P物质免疫阳性神经纤维在各组中的变化:正常对照组(35.50±6.4)%,非治疗组(67.00±3.3)%,治疗组(51.70±4.0)%。非治疗组背根神经节中,患侧P物质免疫反应阳性纤维均较正常组染色明显加深(LSD-t=9.320,P<0.01),而治疗组较非治疗组染色变浅(LSD-t=6.915,P<0.01),但仍较正常组加深(LSD-t=5.177,P<0.01)。结论:提示肢体负压疗法可减少肢体动脉闭塞性病变伤害性刺激的传入,缓解肢体疼痛。AIM:To observe the effect of limb negative pressure(LNP)on substance P(SP) positive immunoreactive nerve fibers in the dorsal root ganglion(DRG) of peripheral arterial occlusive disease in dogs. METHODS: Seventeen dogs were randomly divided into treatment group(n=10),non treatment group(n=5) and normal control group(n=2). The left hindlimb ischemia model was made in the non treatment and treatment groups. The treatment group was treated with LNP for 10 days(15 minutes per day)14 days after the models were made.However,the non treatment group was not treated with LNP.All the groups were stained with immunohistochemical method and the SP positive immunoreactive nerve fibers in the DRG were detected. RESULTS:Alterations of SP positive immunoreactive nerve fibers in the DRG: the normal control group was(35.50±6.4)%,the non treatment group was(67.00±3.3)%,and the treatment group was(51.70±4.0)%.In the DRG in the non treatment group, the staining of SP positive immunoreactive nerve fibers in the suffering side was deeper than that of the normal control group(LSD t=9.320,P< 0.01) and the treatment group was lighter than that of the non treatment group(LSD t=6.915,P< 0.01), however, it was still deeper than that of the normal group(LSD t=5.177,P< 0.01). CONCLUSION:LNP can reduce the transmission of the harmful stimuli from the peripheral arterial occlusive disease and release the pain of limbs.
