痛风灵方对尿酸钠大鼠模型关节软组织细胞间黏附分子1表达的影响(英文)

摘　　要：背景:细胞间粘附分子-1(ICAM-1)在免疫监督、炎症反应、吞噬等过程中起重要作用,但痛风的受损伤组织部位细胞表面粘附分子如何表达,国内外报道较少。目的:为了探讨痛风灵方治疗痛风的机制,在本实验中,研究了痛风灵方对尿酸钠大鼠模型关节软组织细胞间粘附分子1(intercellularadhe-sionmolecule-1,ICAM-1)表达的影响。设计:随机对照的实验研究。地点和材料:实验地点为成都中医药大学。动物:SD健康全雄性大白鼠50只,体质量180～220g,主要药物:痛风灵方由丹参、延胡索、川芎、车前子、泽泻等组成。中药高剂量、低剂量每毫升药液分别含生药3g,1.5g。干预:大白鼠50只按抽签法随机分为阴性对照组、模型组、西药组、中药高剂量组、中药低剂量组,每组各10只。制备尿酸钠溶液及动物造模。阴性对照组、模型组灌胃生理盐水,西药组灌胃萘普生溶液,中药高剂量组、中药低剂量组分别灌胃痛风灵方药液,除阴性对照组不造模外,其余4组均造模。主要观察指标:痛风灵方对ICAM-1的影响。结果:阴性对照组、西药组、中药高剂量组、中药低剂量组的关节软组织ICAM-1水平犤面积总和分别为(178.98±85.22),(250.25±73.44),(188.41±67.09),(191.13±31.39)×103μm;积分光密度总和分别2为(284.17±51.66),(479.27±121.92),(325.96?BACKGROUND:Intercellular adhesion molecule 1(ICAM 1) plays an important role in immune surveillance,inflammatory reaction and phagocytose.There are few reports on how cell surface adhesion molecules express in the injured tissues of gout at home and aboard. OBJECTIVE:To probe into the mechanism of tongfeng ling recipe(Recipe) in treatment of gout and study the effect of tongfeng ling recipe on expression of ICAM 1 in joint tissues in sodium urate rat model DESIGN:Randomized,controlled trial design. SETTING and MATERIALS:The experiment was performed in Chengdu University of Chinese medicine.Firty normal SD male white rats were employed weighted varied from 180g to 220g.The main medicine was tongfeng ling recipe,composed of danshen,yanhusuo,chuanxiong,cheqianzi andzexie.3 g and 1.5 g raw herbs of each milliliter solution were determined as high and low dosages respectively. INTERVENTIONS:According to lot drawing method,fifty rats were divided randomly into negative control group,model group, western medicine group, high dosage of Chinese medicine group and low dosage of Chinese medicine group,ten rats in each group.The animal model and sodium urate solution were self prepared.Physiological saline was for gastric infusion in negative control group and model group,naproxen solution was in western medicine group and the Recipe solution was in high and low dosage of Chinese medicine groups.Except in negative control,the model was made in the rest four groups. MAIN OUTCOME MEASURES:Effects of the Recipe on ICAM 1. RESULTS:Compared with model group [the total square of ICAM 1 was(746.72±112.65)×103μm2,and integrated absorbance(A)was(1 358.62 ±184.66)×105],there was great significantly differences(t=4.211-4.725,P < 0.01) in ICAM 1 level of joint soft tissues in negative control group,western medicine group,high dosage of Chinese medicine group and low dosage of Chinese medicine group[the total squares of ICAM 1 were(178.98±85.22),(250.25±73.44),(188.41±67.09),(191.13±31.39) ×103 μm2 successively
