摘 要:目的 :研究利水通络口服液对大鼠急性脑缺血损伤的影响及其作用机理。方法 :以急性不完全性脑缺血大鼠为模型 ,将5 0只普通级Wistar雄性大鼠随机分为假手术组、模型组、利水通络口服液高剂量组、利水通络口服液低剂量组 ,分别灌胃取材后 ,测定大鼠血清中IL - 8、TNF -α含量 ,脑含水量及脑组织病理形态学变化。结果 :①急性脑缺血后 ,模型组大鼠血清中IL -8、TNF -α含量均明显高于假手术组 (P <0 .0 5 ) ,利水通络口服液高、低剂量组均能降低大鼠血清中IL - 8、TNF -α含量 (P <0 .0 5 )。②急性脑缺血后脑组织中含水量均急剧升高 ,利水通络口服液高、低剂量组在降低脑含水量方面 ,与模型组相比 ,效果显著 (P <0 .0 5 )。③病理组织学观察结果表明 ,急性脑缺血模型组出现了较严重的脑组织水肿和神经细胞病变 ,利水通络口服液高、低剂量组在改善脑组织水肿和神经细胞病变方面效果显著 (P <0 .0 5 )。结论 :急性脑缺血大鼠脑组织病理形态改变及含水量升高均明显 ,血清中IL - 8、TNF -α含量也升高 ,这些变化是脑梗塞急性期的病理生理基础之一 ;利水通络口服液对急性脑缺血大鼠脑组织病理形态有较强的改善作用 ,其作用机制可能与降低含水量有关。Objective: To investigate the effect of Lishui Tongluo Capsule(LSTLC) on the da mage of rats with acute cerebral ischemic and its effective mechanism.Methods: The rats with acute whole-cerebral ischemia were used as models. 50 common male W istar rats were randomly divided into sham-operation group, model group, and hig h-dosed,low-dosed groups of LSTLC. After perfusing stomach and obtaining tissues ,we measurate the levels of IL-8,TNF-α,and water in the cerebric tissues, and o bserve the pathomorphologic changes with optical microscope.Results:① The conte nts of IL-8,TNF-αin the serum of model group increased greatly than sham-operat ion group(P<0.05) after acute cerebral ischemia.The levels of IL-8,TNF-αred uce in the two LSTLC groups (P<0.05). the high levels of IL-8,TNF-αwere dec reased in the high-dosed of LSTLC group (P<0.05).It was remarkable in low-do sed LSTLC group too(P<0.05).② The content of water increased evidently in t he cerebric tissues after acute cerebral ischemia.Compared with model group, the cure effects were remarkable on decreasing the levels of water in high-dosed of LSTLC group (P<0.05). It was remarkable in low-dosed LSTLC group too(P< 0.05).③ Histopathological investigation showed that there were serious cerebric edema and pathologic changes of nerve cells in acute cerebral ischemic model gr oup.High-dosed and low-dosed groups of LSTLC were remarkable in improving cerebr al mydrops and pathologic changes of nerve cells which were induced by acute cer ebral ischemia.Conclusions:Obvious pathomorphologic changes and the high content s of IL-8,TNF-αin the serum and water in cerebric tissues with acute cerebral i schemic are pathological and physiological basis of acute ischemic apoplexy. LST LC can greatly improve the pathomorphology of the rats cerebral tissues with acu te cerebric ischemia.Its mechanism of action may be related to reducing the cont ents of water in cerebric tissues.
